利用植物农药残渣生产绿色木霉孢子的研究

摘要:
[目的]研究利用植物农药残渣作为生产绿色木霉(Trichoderma viride)制剂培养基质的可行性.[方法]利用植物农药沙地柏、川楝和雷公藤的残渣作为培养基主要成分,采用平皿筛选法筛选出最佳组合,然后利用固体发酵法生产绿色木霉孢子制剂.[结果]沙地柏:麦麸:鸡粪:硫酸铵的最佳质量组合比例为12:4:3:1.利用该组合进行28℃恒温培养箱固体发酵培养,绿色木霉产孢量第8天达到最高,沙地柏、雷公藤和川楝残渣的产孢量分别为4.87×107,1.43×108和8.45×107g-1.利用沙地柏残渣进行大体量室温固体发酵,第9天产孢量为2.9×107g-1,第12天产孢量达到最高,为3.6×108g-1,最适培养时间为8~12 d.固体发酵生产的两种孢子有着明显的分布规律,分生孢子主要分布在有光照的表层,厚垣孢子则分布在培养基质的深层.[结论]植物农药沙地柏、雷公藤和川楝的残渣可以用作生产绿色木霉孢子的培养基质.

Study on producing spores of Trichoderma viride with plant-pesticide residues
Abstract:
【Objective】 The research was to study the feasibility of producing spore preparation of Trichoderma viride with plant-pesticide residues.【Method】 The residues of Sabina vulgaris,Tripterygium wilfordii and Melia toosendan were used as major components of the medium for plate screening test and solid fermentation,mixed with some other materials.【Result】 The orthogonal combination experiment indicated that the optimal mix of S.vulgaris,wheat bran,chook dung and ammonium sulfate was 12∶4∶3∶1.The spore quantity of T.viride grown in the solid fermentation media made with S.vulgaris,T wilfordii and M.toosendan respectively achieved the highest on 8th day and the spore densities were 4.87×107,1.43×108 and 8.45×107g-1 respectively.During the solid fermentation with the S.vulgaris mixes,spore density was 2.9×107g-1 on the 9th day and achieved the peak on the 12th day,3.6×108g-1.It was found that there were two kinds of spores in the media fermented,conidium mainly distributed in the surface layer,but chlamydospore distributed in the culture medium in-depth.【Conclusion】 The results showed that the plant-pesticide residuces such as S.vulgaris,T.wilfordii and M.toosendan mixed with some other materials can be used as amedium to produce spore preparation of T.viride.
