中国荷斯坦奶牛LOX-1基因3′UTR的PCR-SSCP分析

摘要:
为了寻找影响奶牛产奶性状的候选基因,采用PCR-SSCP分子标记方法,对中国荷斯坦奶牛LOX-1基因3′非编码区进行了多态性分析。结果表明,在中国荷斯坦奶牛中检测的LOX-1基因3′UTR有AA型、BB型和AB型3种基因型,其频率分别为0.137 4,0.313 0和0.549 6;不同基因型与中国荷斯坦奶牛的乳脂率之间有显著的相关性(P<0.05),但对乳蛋白率、产奶量和体细胞评分(SCS)影响不大,未达到显著水平。初步认为LOX-1基因可能是控制中国荷斯坦奶牛产奶性能的一个候选基因,或是与之存在紧密遗传连锁的分子标记。

Study on the 3′UTR of LOX-1 gene in Chinese Holstein by PCR-SSCP
Abstract:
In order to detect polymorphisms in the lectin like oxidized low density lipoprotein receptor 1(LOX-1) gene,the polymorphism of LOX-1’s 3′UTR in Chinese Holstein was detected through PCR-SSCP (Polymerase Chain Reaction Single Strand Conformation Polymorphism).The result showed that three genotypes AA,BB and AB of the 3′UTR of LOX-1 gene were found,and the genotype frequency were 0.137 4,0.313 0,0.549 6,respectively in Chinese Holstein.There was highly significant correlation between different genotypes in fat content(P<0.05).No significant effect of different genotypes on their ME,protein content and SCS (Somatic Cell Score) was found.These results preliminarily showed that the LOX-1 gene was either a candidate gene that influenced the prolificacy in Chinese Holstein or a molecular marker in close linkage with such a gene.
