5个山羊品种GH基因第Ⅱ、Ⅴ外显子的多态性及其应用

摘要:
为了将GH基因应用于山羊标记辅助选择中,利用PCR-SSCP技术分析了西农萨能奶山羊、关中奶山羊、陕北白山羊、安哥拉山羊和波尔山羊5个山羊品种生长激素(gGH)基因外显子Ⅱ、Ⅴ的多态性及其与西农萨能奶山羊生产性能的关联。结果表明,gGH基因外显子Ⅱ在西农萨能奶山羊和关中奶山羊群体中存在多态性,外显子Ⅴ在除波尔山羊外的其他山羊品种中均存在多态性;品种间gGH基因外显子Ⅴ位点的基因型频率、基因频率分布与品种特性显著相关(P<0.05);gGH基因外显子Ⅱ的多态性与产羔数之间存在显著关联(P<0.05),外显子Ⅴ的多态性与产奶量之间显著关联(P<0.05)。说明gGH基因位点对产奶量、产羔数有显著影响,gGH基因外显子Ⅱ、Ⅴ的多态性分别可作为产羔数、产奶量标记辅助选择的DNA标记。

Polymorphisms of exon Ⅱ and Ⅴ of GH gene in five goat breeds and their application in production traits
Abstract:
In order to contribute to increase the possibility of application of GH gene in MAS and improve goat breed,PCR-SSCP was applied to analyze polymorphisms of exon Ⅱ and Ⅴ for GH gene in five goat breeds,namely,Xinong Sannen dairy,Guanzhong dairy,Shaanan White,Angora and Boer,and their associations with production traits of Xinong Saanen dairy goat.The results showed that there were polymorphisms in exon Ⅱ locus in Xinong Saanen dairy and Guanzhong dairy populations,and there were polymorphisms in exon Ⅴ loci among Xinong Sannen dairy,Guanzhong dairy,Shaanan White,and Angora populations;distribution of genotype frequencies and gene frequencies were significantly associated with breed factor in exon Ⅴ loci (P<0.05 or P<0.01).Moreover,significant statistical correlations between milk yield and exon Ⅴ polymorphism was found in Xinong Saanen dairy goat (P<0.05);association of polymorphisms of exon Ⅱ with litter sizes was significantly found in Xinong Sannen dairy goat (P<0.05).The study confirmed that the exon Ⅴ an exon Ⅱ had positive effect on milk yield,and litter size,which could be suggested for perfect molecular marker in marker-assisted section (MAS).
