葡萄风信子基因转化受体系统的建立

摘要:
以葡萄风信子(Muscari botryoides Mill)品种“紫葡萄”(Cantat)的鳞茎和叶片为外植体,通过植物组织培养再生系统的建立和抗生素敏感性试验,建立了稳定的葡萄风信子基因转化受体系统。结果表明,葡萄风信子的鳞茎和叶片均可被诱导形成大量的愈伤组织;叶片诱导愈伤组织的最适培养基为MS+0.6 mg/L 2,4-D+0.1 mg/L 6-BA,其诱导频率可达100%;鳞茎诱导愈伤组织的最适培养基为MS+3.0 mg/L 2,4-D +0.3 mg/L 6-BA,其诱导频率达91.2%;愈伤组织分化不定芽的最适培养基为MS+2.0 mg/L 6-BA+0.1 mg/L NAA,生根最适培养基为1/2 MS+1.0 mg/L 6-BA 0.1%AC;筛选卡那霉素的临界质量浓度为60 mg/L,羧苄青霉素的适宜质量浓度为300 mg/L。

Construction of gene transformation acceptor system in Muscari botryoides
Abstract:
In this study,bulbs and leaves of grape hyacinthus orientalis “Cantat” were used for explants,a high-frequency regeneration system by plant tissue culture was established,the experiment about antibiotic sensitivities were conducted,and a stably-performing and high-efficiency acceptor system for the gene transformation of grape hyacinthus orientalis leaves were established.The results showed abundant callus could be induced from the bulbs and leaves of grape hyacinthus orientalis explants.The optimum medium for inducing callus from leaves was MS+0.6 mg/L 2,4-D+0.1mg/L 6-BA,and its induction frequency could reach 100%;the combination of MS+3.0 mg/L 2,4-D+0.3 mg/L 6-BA was the appropriate medium for bulbs forming callus with the induction rate 91.2%.The best medium for inducing adventitious buds from callus was MS+2.0 mg/L 6-BA+0.1 mg/L NAA.The medium of 1/2 MS+1.0 mg/L 6-BA+0.1% AC was suitable for rooting.What's more,the concentrations of the antibiotics of test-tube plantlets in their cultures were also determined;the quality concentration of kanamycin was 60 mg/L and the quality concentration of carbenicillin was 300 mg/L.
