皱柄白马鞍菌液体摇瓶培养条件优化

摘要:
通过摇瓶液体培养试验,对皱柄白马鞍菌液体深层培养中培养基成分、pH值、装液量及摇床转速、培养时间等对菌丝体生长的影响进行了对比研究。结果表明,适宜的培养条件为:培养基含小麦粉20 g/L、硫酸氨2 g/L、MgSO4 0.5 g/L、KH2PO4 0.46 g/L、K2HPO4 1 g/L,最适pH 5.5-6.5,装液量300~400 mL/L,接种量5 mL培养菌液,温度26-28℃;摇床转速100 r/min,培养时间7 d。适宜条件下最大菌丝生物量可达5.867 g/L。

Optimization of liquid submerged culture conditions for the mycelial growth of Helvella crispa Fr.
Abstract:
The culture conditions of Helvella crispa Fr.in shake flask liquid culture were investigated in the experiment.Results showed that the optimal medium of liquid submerged culture was as follows:wheat flour 20 g/L,HN4SO4 2 g/L,MgSO4 0.5 g/L,KH2PO4 0.46 g/L,K2HPO4 1.0 g/L;when the medium capacity was 300-400 mL/L,and the seed (about 5 mL) was inoculated,the shake flask culture experiment was carried out at 26-28 ℃,pH 5.5-6.5 in a shake incubator with a rotation rate 100 r/min for 7 days.The maximum mycelial biomass was as high as 5.867 g/L.
