| 哺乳动物有腔卵泡中的卵母细胞在体外会自发恢复减数分裂,适当延迟减数分裂的自发恢复,能促进核与胞质成熟的同步化,有利于后续胚胎的发育。本试验研究了磷酸二酯酶3(PDE 3)特异性抑制剂milrinone对山羊卵母细胞体外成熟及孤雌胚发育的影响。结果表明,milrinone抑制山羊卵母细胞体外成熟的时间为6 h,其最佳剂量为100μmol/L,经milrinone处理体外培养28 h成熟的山羊卵母细胞用于孤雌激活,能显著提高孤雌激活胚的卵裂率及囊胚率。说明成熟培养液中添加适当剂量的milrinone,能显著提高体外成熟的山羊卵母细胞的质量。 |

| The effects of phosphodiesterase inhibitors on maturation and parthenogenesis of goat oocytes in vitro |

| Abstract: |
| The oocytes of antral follicles can spontaneously resume meiosis in vitro in mammalian,properly delay the process of spontaneous meiotic maturation in vitro may promote the homochronous maturation of nucleus and ooplasm and benefit the following embryos development.The experiment researches the effects of phosphodiesterase type 3 (PDE 3) inhibitors milrinone on maturation and parthenogenesis of goat oocytes in vitro.The results show that the time of the PDE 3 inhibitor delaying the goat oocyte mature in vitro is 6 hours,the best dosage is 100 μmol/L.After parthenogenetic activation,the goat oocytes after 28 hour matured culture in PDE 3 inhibitor-supplemented media have both higher cleavage rate and blastocyst rate.Therefore,milrinone supplemented media can significantly enhance the quality of goat oocyte matured in vitro. |
