利用RAPD分子标记技术,从400条10碱基的随机引物中筛选出了40条适用于枸杞RAPD分析的引物,选取其中9条引物对10份不同来源的宁夏枸杞材料进行了扩增,检测了宁夏不同地区种植的宁夏枸杞(Lycium barbarum L.)主栽品种和新育成的枸杞品系基因组DNA的多态性,并对其遗传背景进行了初步分析。结果表明,宁夏不同地区主要栽培的宁夏枸杞品种遗传上无明显差异,但新品系大果枸杞与标准宁杞1号在基因组上有差异;同时构建了主要推广品种宁杞1号的RAPD图谱。 |

RAPD analysis of Lycium barbarum L.cultivated in diferent areas of Ningxia |

Abstract: |
RAPD (random amplified polymorphic DNA) was used to appraise the genetic diversity of Lycium barbarum L.gathered from different areas of Ningxia.40 oligonucleotide primers from 400 oligonucleotide primers were selected for the RAPD analysis.The 10 DNA samples from diferent places were amplified by 9 selected primers.The result indicates that there are no significant genetic diferences among the 10 DNA samples,but the new bred material shows several different strips from the main variety Ningqi No.1.And DNA fingerprint of Ningqi No.1 was built by the RAPD techniques. |
