粘虫细胞培养及苦皮藤素Ⅳ,Ⅴ对其毒力的研究

摘要:
以东方粘虫(Mythimna separate Walker)为材料建立了新的昆虫细胞系,原代培养6个月后,胚胎细胞开始传代;用苦皮藤素和处理第3代传代细胞,24 h后细胞裂解,出现大量细胞碎片,培养液变浑浊,其致死中质量浓度分别为76.42和129.34 μg/mL。粘虫背纵肌经胰蛋白酶部分消化后在Grace培养液中生长良好,培养2个月后有大量肌卫星细胞出现。

The cell culture of Mythimna separate and cytoxicity assay of Celangulin Ⅳ and Ⅴ against the cell
Abstract:
After 6 months’primary culture,Mythimna separate Walker embryo cell began continuous culture,and the 3rd generation of continuous culture cells was treated by Celangulin Ⅳ and Celangulin Ⅴ.The result showed that lysated after 24 hours,the medium became haze,and the value of LC50 were 76.42 and 129.34 μg/mL respectively.In addition,Primary cultures were done in Grace medium after the middle dorsal vertical muscles from female adult Mythimna separate Walker was partly digested by enzyme,and the growth of Myoblast cells cultured was better and clear;after 2 months’culture,large number of satellite cells appeared.
