苦皮藤内生真菌Hd3菌株的研究

摘要:
从新鲜苦皮藤根韧皮部分离筛选到1株内生真菌,编号为Hd3。生测结果表明,当质量浓度为50.0 mg/mL时,Hd3菌株菌丝乙酸乙酯提取物对3龄粘虫、小菜蛾的胃毒活性分别为100%和26.67%;盆栽法处理中,当质量浓度为44.0 mg/mL时,Hd3菌株菌丝乙酸乙酯提取物对小麦蚜虫的杀虫活性为94.11%。选用硅胶柱层析和反相半制备高效液相色谱切割分离,从Hd3菌株菌丝体乙酸乙酯提取物中得到1个杀虫活性物质H4.5,经波谱数据分析,化合物H4.5的结构为2,3-二甲氧基-5-甲基苯酚。通过形态特征、培养特性等方面的研究,将Hd3菌株初步鉴定为曲霉属(Aspergillus spp.)环绕亚属(Subgen.Girumdati)真菌。

Studies on the endophytic fungus Hd3 strain of Celastrus angulatus
Abstract:
Endophytic fungus Hd3 strain was isolated from the phloem of the root of Celastrus angulatus.The results of bioassay indicated the ethyl acetate extract of the endophytic fungus Hd3 strain mycelium had stomach toxicity against the 3rd instar larvae of Mythimna separate and Plutella xylostella at the concentration of 50.0 mg/mL,and the mortalities were 100 and 26.67 respectively.The extract had a mortality of 94.11 against Macrosiphum avenae at the concentration of 44.0 mg/mL in the pot test.An activity compound H4.5 was isolated from the extracts by p-TLC and HPLC.On the basis of spectral technology 1H NMR,13 C NMR and MS,the structure of H4.5 was identified as 2,3-dimethoxy-5-methyl phenol.Based on its morphological,cultivation and physiological characteristics,strain Hd3 was identified as Aspergillus spp.
