高压静电场对老化水稻种子幼苗根系的影响

摘要:
将吸胀萌动后的老化水稻种子随机分为Ⅰ~Ⅴ 5个处理组,分别用250,300,350,400,450 kV/m的高压静电场处理55 min,然后30 ℃浸种催芽24 h,另外设CK1(未老化水稻种子)和CK2(老化水稻种子)2个对照,研究高压静电场对老化水稻种子发芽第4天幼苗根系生长发育的影响。结果表明,适宜的高压静电场处理组幼苗的根系活力、可溶性蛋白质含量均极显著高于对照组(P<0.01);与其他处理组相比,Ⅲ,Ⅳ,Ⅴ处理组蛋白质电泳条带数及条带颜色加深,说明这3个处理组可能影响了老化水稻种子根系中部分蛋白质的表达;与对照相比,Ⅱ~Ⅴ高压静电场处理组有丝分裂指数极显著增加,但未见染色体畸变。可见高压静电场处理不仅促进了老化水稻种子幼苗根系的分裂生长,同时还为其生长提供了充足的营养。

Effect of high voltage electrostatic field on the sprout roots of aging rice seeds
Abstract:
In this paper the effect of HVEF on the growth and development of rice sprout roots on the fourth day of germination was studied.The wet aging rice seeds were divided into five groups (Ⅰ-Ⅴ) stochastically,and were treated by HVEF with doses 250,300,350,400,450 kV/m for 55min,then the seeds were dipped for 24 h at 30 ℃,and with CK-1 which was not aging seeds and CK2 which was aging seeds as control groups.The results showed that in the appropriate treated groups,the root vigor,the content of soluble protein of roots were significantly higher than those of the control groups (P<0.01).The numbers of piece and colour of electrophoresis map in Ⅲ,Ⅳ,Ⅴ groups were more enhancive than those of other groups.So the expression of the roots-partial soluble protein in the Ⅲ-Ⅴ groups was possibly affected by the treatment.The mitotic index of root tip cells in Ⅱ-Ⅴ groups was significantly higher than that of the control groups,and without the aberrance of chromosome in treatment groups.So HVEF not only promoted the cell division and the growth of aging rice sprout roots,but supplied ample nourishment for its growth.
