氰戊菊酯对Trichoplusia ni细胞活力的影响

摘要:
以不同浓度的氰傥菊酯处理昆虫细胞Trichoplusia ni(Tn),并用台盼兰染色、3(4,5-Dimethylthiazol)-2,5-diphenyltrazolium bromide(MTT)还原试验及流式细胞仪检测等方法对Tn细胞活力进行测定。结果显示,氰戊菊酯浓度大于17.5 μmol/L处理的Tn细胞形态、活力均发生了明昆改变。在台盼兰染色实验中,氰戊菊酯浓度≥17.5 μmol/L时,Tn细胞死亡率升高到29.69%以上,极显著高于对照(P<0.01);在MTT还原实验中,当氰戊菊酯浓度≥17.5 μmol/L时,Tn细胞对MTT试剂的还原物吸光值降低,细胞相对活力较对照极显著下降(P<0.01);用流式细胞仪检测的结果也证实,氰戊菊酯浓度为25 μmol/L时,死亡与凋亡细胞数明显高于对照。

Vitality of Trichoplusia ni(Tn) cell treated with fenvalerate
Abstract:
The Trichoplusia ni(Tn) cells were treated with fenvalerate at ladder concentrations and the vitalities of the cells were detected by Typan blue dye exclusion,MTT reduction assay and Flow cytometer (FCM).Results show that the vitalities of fenvalerate-treated Tn cells were lower than those of untreated cells significantly when the concentrations arrived at 17.5,20 and 25 μmol/L (P<0.01) in Typan blue dye exclusion and MTT reduction assay.The death and apoptosis cells were increased notably between the cells treated with 25 μmol/L fenvalerate and control.Those results could be a basic study for further research of mechanism of detoxified enzyme induced by fenvlerate.
