陕西线辣椒种子携带和传播病毒研究初报

摘要:
2004-12~2005-01,从陕西省线辣椒主产区随机采集3个线辣椒品种的28份种子样品,经酶联免疫吸附法(ELISA)和RT-PCR法检测发现,28份种子样品中,与烟草花叶病毒(TMV)抗血清呈阳性反应者占14.29%,呈弱阳性或可疑反应者占42.86%;与黄瓜花叶病毒(CMV)抗血清呈阳性反应者占7.14%,呈弱阳性或可疑反应者占46.43%;仅有1份种子(占3.57%)与马铃薯Y病毒(PVY)抗血清出现弱阳性或可疑反应,其余均为阴性反应。线辣椒苗期生长观察试验表明,只有对TMV表现强阳性反应的3份种子样品,在出苗后40 d出现轻型花叶症状,其他对TMV,CMV和PVY表现弱阳性和阴性反应的样品均未出现可见症状,直至出苗后70 d依然如此,说明种子带毒量较低时不足以引起幼苗发病。RT—PCR检测结果表明,随机抽取的5份样品播种后,均未从其幼苗的叶片中扩增出蚕甄萎蔫病毒(BBWV)的CP基因,说明这5份种子样品的幼苗中均不存在BBWV。研究结果还表明,采自宝鸡地区的种子样品TMV带毒率和带毒量较高;采自咸阳和渭南地区的种子样品CMV带毒率较高.但带毒域较低;采自宝鸡地区风翔县的3份样品3种病毒均未检出;不同品种种子带毒率无明显变化规律。

Preliminary report on transmission of viruses through line pepper (Capsicum annuum L.) seed in Shaanxi province
Abstract:
To understand the transmission of virus through line pepper seed and supply evidence for seed treatment and production,the TMV,CMV,PVY and BBWV on 28 seed samples,which were collected from 3 major line pepper production areas of Shaanxi province from December of 2004 to January of 2005,of 3 line pepper cultivars were examined by ELISA and RT-PCR respectively.The results of ELISA indicated that:14.29% of samples showed strong positive reaction and 42.86% showed weak positive reaction to TMV;7.14% of samples showed positive reaction and 46.43% showed weak positive to CMV;only one sample showed weak positive reaction to PVY.In seedling stage,3 samples that showed strong positive reaction to TMV performed the symptom of TMV on the 40th day after sprouting,and the other seedlings were normal until the 70th day after sprouting.These results indicated that the viruses in most samples were not strong enough to cause the virus disease in seedling although some of them showed weak positive reaction to virus.The seedlings of 5 samples,which were selected randomly from 28 samples,were examined by RT-PCR.The results showed that the PC gene of BBWV was not amplified by means of RT-PCR and there wasn't BBWV in the seedlings of these 5 seed samples.The results of this experiment also indicated that the carrying percentage of TMV in samples collected from Baoji area was higher;and the carrying percentage of CMV in samples collected from Xianyang and Weinan area was higher;the 3 samples collected from Fengxiang county were virus free;the percentage of virus in samples was not varied with cultivars.
