| 以胎鼠的神经干细胞为研究对象,将其分别置于FHL,FH,FL,LH 4种培养液中,于37 ℃、50 mL/L CO2、饱和湿度下培养,同时作空白对照试验;7 d后,检测胆碱乙酰基转移酶(ChAT)免疫反应,研究在不同培养液中,神经干细胞向胆碱能神经元定向分化的能力。结果表明,在FHL和FH培养液培养的细胞中,能检测到ChAT阳性细胞,说明FHL和FH培养液可诱导神经干细胞分化为胆碱能神经元。 |

| Study on NSCs being primed and differentiated to cholinergic neurons |

| Abstract: |
| The subject of this research is the neural stem cells obtained from the embryonic mice which are cultured in four different kinds of mediums,37 ℃ and 50 mL/L CO2 saturated humidity.The mediums consist of four different arrays composed of 20 ng/mL bFGF,5 μg/mL Heparin and 1 μg/mL Laminin,then each array is added F12-DMEM (1∶1),including 1% B27 (V/V) and 20 ng/mL EGF.After 7 days of culturing,cells are immunized with ChAT.Results show that ChAT can only be detected in two mediums containing 20 ng/mL bFGF,5 μg/mL heparin,1 μg/mL laminin (FHL) and 20 ng/mL bFGF,5 μg/mL heparin (FH).This result suggests that cholinergic neuron can be primed and differentiated in mediums that contain FHL and FH,which will be important in the treatment of old age dementia and epilepsy. |
