| 设置养分贮存损失状态不同的4类培养液,分别添加一定浓度梯度的L 谷氨酰胺(L-Glutamine,L-Gln)培养大鼠前体脂肪细胞。通过形态学观察、MTT比色、油红O染色提取法,比较各组细胞形态及增殖与分化的效果。结果表明,添加0 mmol/L L-Gln的新鲜培养液和配制后4 ℃冷藏20 d,再添加0.685 mmol/L L-Gln的培养液,均较利于前体脂肪细胞的增殖与分化;配制后4 ℃冷藏90 d的培养液,对前体脂肪细胞培养效果较差,添加L-Gln也不能使培养效果得到改善。另外,启封后的M199和胎牛血清(Fetal Calf Serum,FCS)分别冷藏不宜超过90 d。说明及时满足培养液中L-Gln的需求量,可使前体脂肪细胞达到较好的增殖与分化状态。 |

| Effect of L-Gln on proliferation and differentiation of rat preadipocytes |

| L-Gln was added to the 4 culture media of different nutritions for rat preadipocytes.The proliferation and differentiation of preadipocyte were determined by MTT spectrophotometry and Oil Red O staining respectively.The results show that the addition of 0 mmol/L L-Gln in fresh culture media and 0.685 mmol/L L-Gln in the culture media which had been stored at 4 ℃ for 20 d benefited proliforation and differentiation of the preadipocytes.When culture media was stored at 4 ℃ for 90 d,it could not make the preadipocytes develop soundly,even when L-Gln was added.Once more,M199 and FCS (fetal calf serum) could not be stored at 4 ℃ beyond 90 d if they were opened.The above shows that suitable supplement of L-Gln could make preadipocytes approach better states of proliferation and differention. |
