荣昌猪TYR基因的克隆测序及多态性检测

摘要:
根据人、猪、牛的酪氨酸酶(Tyrosinase,TYR)基因部分序列(GenBank:AF237808,AF237809,AF181962,AY046527),利用Oligo6.0软件设计5对引物,对荣昌猪的TYR基因进行了PCR扩增,并用单链构象多态分析(SSCP)方法进行了多态性检测。结果发现,外显子1呈多态,其余3个外显子均呈单态。克隆测序首次得到猪的酪氨酸酶基因外显子2,3,4的部分序列(序列已被GenBank收录,登录号分别为AY236997,AY279998,AY242973),而且发现外显子1的多态是由于起始密码子上游第46位的G突变成A造成的。

Cloning,sequencing and polymorphism detecting of tyrosinase gene in Rongchang pig
Abstract:
According to known DNA sequences of tyrosinase in human,pig and cattle,we designed five pairs of primers and amplified all exon DNA sequences of tyrosinase in pigs except exon 5.With the method of SSCP,we detected all genotypes in Rongchang pigs.The result revealed exon 1 was polymorphic and other three exons were monomorphic.The products were cloned and sequenced.Three new sequences have been authorized three accession codes:AY236997,AY279998,AY242973.We also found that the polymorphism in exon 1 of tyrosinase gene came from a single nucleotide:G→A.
