化学诱变剂EMS对小麦条锈菌夏孢子萌发率的影响

摘要:
采用3个pH值缓冲液,4个浓度甲基磺酸乙酯(EMS)溶液及6个时间处理,对小麦条锈菌夏孢子的诱变条件进行优化。结果表明,在室温条件下缓冲液pH值为7.0时,经0.03 mol/L EMS溶液处理6~8 min,小麦条锈菌条中27号小种夏孢子的死亡率达83.6%~85.8%,符合微生物诱变经验指数最佳诱变剂量的选择标准。不同毒性的小麦条锈菌生理小种对EMS的敏感性不同,且在离体培养条件下,毒性较弱小种的生活能力及抗逆能力明显强于毒性较强的小种。

Influence of chemical mutagen ethyl methyl sulfomar (EMS) on germination rate of wheat stripe rust (Puccinia striiformis West)
Abstract:
Three pH value of phosphate buffer,four concentrations of EMS solution,and six treatments at different time were used to optimize mutagenic condition of wheat stripe rust urediospore.The results showed that when the pH value was 7.0 in the normal temperature conditions,and uredospores of CY27 disposed for 6-8 minutes with 0.03 mol/L EMS solution,the mortality has reached 83.6% to 85.8%.This result fitted with the selection standards of the optimum mutagenic dose of the microbe chemical mutagenesis.Different rust physiological race has the different sensitivity to the EMS solution.The survival ability of low virulence races were stronger than that of the high virulence in in vitro condition.
