抗TMV的植物筛选及提取条件对抗病毒物质活性的影响

摘要:
对26种植物样品进行了抗病毒(TMV)活性测定。结果表明,不同植物提取物对TMV侵染心叶烟的抑制效果不同,其中莲、榕树、柿子、杨梅、水蜈蚣、叶下珠、羊蹄甲和菊花对TMV的抑制率在90%以上;对高活性抗病毒植物莲叶而言,抗病毒物质的活性因提取溶剂和提取方法的不同而异,水作为溶剂提取时,提取物对TMV的抑制率仅约为70%,而以甲醇和体积分数95%的乙醇作溶剂提取时,提取物对TMV的抑制率约在98%以上;用体积分数95%的乙醇为溶剂,采用索氏提取、回流提取和冷浸提取时,所得提取物对TMV的抑制率均为100%。冷浸提取的优化条件是:以体积分数95%的乙醇为提取溶剂,料液比20:180(1:9)提取12 h。

Screening of plants with resistance to TMV and the influence of extracting condition on the activity of antiviral substance
Abstract:
The antiviral activities were determined by extracts from 26 plants.The results showed that:① Different plant extracts have different inhibitory effect on TMV infecting Nicotiana glutinosa ;Among them,inhibition rates on TMV of Ficus microcarpal,Nelumbo nucifera Gaertu,Myreca rubra Sieb.et Zucc,Diospyros kaki,Kyuinga breviforlia,Phyllanthus urinaria,Bauhinia varitagata and Chrysanthemum morifolium extracts are all over 90%;② As for lotus leaves,the antiviral substance activity is dissimilar based on extracts from lotus leaves with different solvent and methods,inhibition rate on TMV of the extract with water is only around 70%,whereas,inhibitory rate of methanol or 95% (V/V) ethanol are over 98%;The inhibition rate on TMV of the extract from lotus leaves with 95%(V/V) ethanol by Soxhlet extractor,reflux and cold soak are all 100%.Optimum condition of the cold soak are:95% (V/V) ethanol is used as solvent,the ration of material to solvent is 20∶180 (1∶9),soak time is 12 h.
