猪H-FABP基因PCR-RFLP分子标记研究

摘要:
利用PCR-RFLP(HinfⅠ、HaeⅢ和MspⅠ 3种限制性内切酶)分子标记技术,检测了杜洛克猪、长白猪、大白猪、内江猪、荣昌猪、汉江黑猪、汉白猪、八眉猪和野猪等8个猪种,共计265头猪心脏脂肪酸结合蛋白基因5′-上游区和第二内含子区的遗传变异。结果表明,在HinfⅠ-RFLP位点上,上述猪种和野猪均存在多态性,等位基因H的频率分别为0.750 0,0.718 8,0.916 7,0.333 3,0.125 0,0.690 9,0.116 7,0.850 0和0.937 5;除汉江黑猪(P<0.05)和野猪(P<0.01)外,其余猪种的基因频率和基因型频率都处于Hardy-Weinberg平衡状态(P>0.05);大白猪、八眉猪、汉江黑猪、汉白猪和野猪表现为低度多态性(PIC<0.25),杜洛克猪、长白猪、内江猪和荣昌猪为中度多态性(0.25<PIC<0.5);而在HaeⅢ-RFLP和MspⅠ-RFLP位点上,仅内江猪、荣昌猪、汉江黑猪和八眉猪为单态。

Abstract:
In this experiment,genetic variations of 5′-upstream region and the second intron in 265 pig H-FABP gene including Duroc,Landrance,Largewhite,Neijiang pig,Rongchang pig,Hanjiang Black pig,Hanzhong White pig,Bamei pig and wild pig were checked by PCR-RFLP Molecular Marker with HinfⅠ,HaeⅢ and MspⅠ.The result showed that for HinfⅠ-RFLP,the above pig breeds had polymorphism and frequencies of allele H are 0.750 0,0.718 8,0.916 7,0.333 3,0.125 0,0.690 9,0.116 7,0.850 0 and 0.937 5 respectively;expect for Hanjiang Black pig(P<0.05) and wild pig (P<0.01),frequencies of gene and genotype in other breeds are in equilibrium of Hardy Weinberg (P>0.05);largewhite,Bamei pig,Hanjiang Black pig,Hanzhong White pig and wild pig present low polymorphism (PIC<0.25) while other breeds have mediat polymorphism(0.25<PIC<0.5).Among tested breeds,only 4 Chinese local pig breeds have no polymorphism at the HaeⅢ-RFLP and MspⅠ-RFLP sites.
