不同直径卵泡对猪卵母细胞体外成熟的影响

摘要:
以TCM-199成熟培养猪卵巢表面直径2~3 mm,3~5 mm和5~8 mm卵泡内的卵母细胞,比较了成熟培养液中添加以上各组卵泡液(pFF)对猪卵母细胞体外成熟(IVM)的影响,以成熟培养48h后排出第一极体为成熟标准对猪卵母细胞进行成熟鉴定。结果表明,2~3 mm,5~8 mm和3~5 mm 3组卵泡内卵母细胞的体外成熟(IVM)率依次升高,分别为12.7%,31.3%和46.0%,各组间差异显著(P<0.05);添加3~5mm卵泡液的卵母细胞体外成熟率(42.0%)也显著高于5~8 mm组(11.0%)和2~3 mm组(12.7%)(P<0.05)。试验同时观察记录了不同直径卵泡内卵母细胞的等级,结果发现5~8mm组中A级卵母细胞的比例(37.7%)显著高于3~5 mm组(23.9%)(P<0.01),但其B级卵母细胞的比例(45.9%)却显著低于2~3 mm(62.1%)组和3~5 mm(59.2%)(P<0.05)。这说明,2~3 mm,3~5 mm和5~8 mm 3组卵泡中,卵泡直径增大,其在猪卵母细胞体外成熟中的优势渐增。

Effect of follicles with different diameters on porcine oocyte in vitro mature
Abstract:
Porcine oocytes from follicles in diameters of 2-3 mm,3-5 mm and 5-8 mm were matured in vitro in TCM-199 and the effects of pFF from the above groups on the in vitro maturation (IVM) during maturing were compared.The matured oocytes were certified if the PbⅠexcluded after 48 hour's maturing.The results were as follows:the in vitro maturation (IVM) rates of porcine oocytes derived from follicles in diameters of 2-3 mm,5-8 mm and 3-5 mm were increased gradually,the rates were 12.7%,31.3% and 46.0% respectively and the difference between them was significant (P<0.05);The rates of matured oocytes with pFF from 3-5 mm group (42.0%) was significantly higher than that from 2-3 mm group (12.7%) and 5-8 mm group (11.0%) (P<0.01);The oocyte classes from different diameters follicles were observed and recorded,the proportion of class A oocytes in 5-8 mm group (37.7%) was significantly higher than that from 3-5 mm group (23.9%),but the proportion of class B in 5-8 mm group was lower than that in 2-3 mm (62.1%) and 3-5 mm (59.2%) (P<0.05).The results indicated that the superiority of porcine follicle in diameters of 2-3 mm,5-8 mm and 3-5 mm increased gradually in vitro maturation of porcine oocytes.
