苹果黑星病菌DNA提取方法研究

摘要:
采用CTAB法、SDS法、Parker法及改进的SDS法提取苹果黑星病菌的DNA,经紫外分光光度计测定,改进SDS法提取的DNA得率较其他3种方法高,约为Parker法的2倍。改进SDS法提取的DNA A260nm/A280nm值为1.700~1.900,DNA纯度较高,而其他3种方法提取的DNA A260nm/A280nm值均高于1.900,DNA中所含RNA的量较高。4种方法提取的DNA在230nm波长处的吸收值分别为0.658,0.257,0.926和0.208,说明DNA不同程度地被酚类物质所污染,但改进的SDS法提取的DNA受污染程度最小。

Study on the method for DNA extraction of Venturia inaequalis
Abstract:
DNA extraction quality of Venturia inaequalis using CTAB method,SDS method,Parker's method and modified SDS method was detected by ultraviolet spectrometer.Results showed that DNA extraction rate using modified SDS method was higher than that of other three methods.It was twice of DNA extraction rate of Parker's method.A260nm/A280nm value of DNA from modified SDS method was between 1.700 and 1.900.It showed that the DNA had high purity.However,A260nm/A280nm value of DNA from other three methods were greater than 1.900.The DNA contained RNA.The abstraction value of 4 methods at 230 nm was 0.658,0.257,0.926 and 0.208 respectively.It showed that DNA was polluted by phenol at different level.Among them,DNA from modified SDS method was polluted slightly.
