硝酸银对中国李原生质体分离和培养的影响

摘要:
以种胚愈伤组织为试材,研究了硝酸银对中国李原生质体分离和培养效果的影响。结果表明,在酶解液中加入4~10 mg/L质量浓度的硝酸银,可提高原生质体的活力,其中以8 mg/L的效果最好,原生质体的活力比对照提高21.1%,但其对原生质体产量无明显作用。在培养基中加入4~8 mg/L质量浓度的硝酸银,原生质体的分裂频率和植板率分别比对照增加19.85%~38.24%和9.23%~36.53%。硝酸银对愈伤组织不定芽的再生能力也具有明显的促进作用。

Effects of silver nitrate on protoplast isolationand culture of Prunus salicina
Abstract:
The effects of silver nitrate on protoplast isolation and culture of Prunus salicina variety “Wanhongli” were studied by using calli derived from embryos.The results showed that the addition of silver nitrate to enzyme solution at the concentration of 4-10 mg/L increased the viability of protoplasts,and the most effective concentration was 8 mg/L.At this concentration,the viability of protoplasts was 21.1% higher than that of control.No effect of silver was found on the yield of protoplasts.The division frequency and plating frequency of protoplasts were increased 19.85%-38.24% and 9.23%-36.53% respectively by adding 4-8 mg/L silver nitrate.Stimulation of shoot regeneration from calli was also found when silver nitrate was added to the culture medium.
