玻璃化冷冻保存牛卵母细胞技术研究

摘要:
采用一步法和二步法研究玻璃化冷冻保存的牛生发泡(GV)期和体外成熟(IVM)卵母细胞的发育潜力。结果表明,冷冻基础液PBS和TCM199在玻璃化冷冻中的效果差异不显著(P>0.05);冷冻程序和卵母细胞发育阶段对冷冻效果有明显影响,二步法冷冻效果显著优于一步法(P<0.05),IVM卵母细胞冷冻效果显著优于GV期(P<0.05)。

Vitrificated cryopreservation of bovine oocytes
Abstract:
The developmental competences of bovine IVM and GV-stage oocytes cryopreserved by vitrification were studied.The results showed that there was no significant differences between TCM199 and PBS in vitrification as the holding medium (P>0.05),whereas the step of cryopreservation and development stage had significant effects on the development capacity of frozen-thawed bovine oocytes,the development capacity with two-step vitrification was better than that with one step vitrification after thawing.The fertilization rates and the cleavage rates of GV oocytes were significantly lower than that of the IVM oocytes(P<0.05).
