生长点点滴法在筛选鉴定转基因植株中的应用

摘要:
对转化体(种子或幼苗)筛选的培养基筛选法和生长点点滴筛选法进行了比较分析。结果表明,在4 d苗期的转化芥菜幼苗生长点点滴800 mg/L的潮霉素,获得了转基因植株,PCR检测结果表明,两种筛选方法的筛选结果一致。生长点点滴滴法的优点是,转基因幼苗不易污染,且成活率高。

Dripping antibiotic solution on growing point as a novel approach to screening transgenic plants
Abstract:
The two methods of screening transgenic plants,including transgenic seeds sowed in the medium with hygromycin (TSSM) and dripped hygromycin solution on the growing point of transgenic seedling (DSGPTS),were compared and analysed.The results indicated that the transgenic plants were obtained with the method of DSGPTS,when 800 mg/L of hygromycin was dripped on the growing point of transgenic seedling at its age of 4 days.PCR analysis showed that the results of both TSSM method and DSGPTS method were showing no difference.The advantage of DSGPTS method is that the transgenic seedling is safe from pollution and the percentage of survival for the transgenic seedling is high.
