分蘖洋葱茎尖培养脱毒苗技术研究

摘要:
利用阿城紫皮分蘖洋葱进行茎尖培养脱毒苗进行研究。结果表明:最佳诱芽培养基为MS+NAA 0.1 mg/L +BA mg/L;最佳生根培养基为1/2MS+NAA 0.01 mg/L +IBA 1.5 mg/L +PP333 0.1 mg/L。以上各培养基均加蔗糖30 g/kg、琼脂8 g/kg、pH值调至5.7~5.8。试验还就外植体的大小、鳞茎热处理对脱毒效果的影响进行了探讨。细胞学观察表明,该途径保持了原材料的遗传稳定性。

Study on virus elimination of tillered-onion by meristem tip culture
Abstract:
The results of virus elimination by shoot- tip culture of purple-skinned tillered-onion commonly grown in Acheng city showed that the best medium for shooting was MS+NAA 0.1 mg/L+BA 0.4 mg/L ,and that for rooting was 1/2 MS+NAA 0.1 mg/L+IBA1.5 mg/L+PP3330.1 mg/L.Each medium was added 30 g/kg sucrose and 8 g/kg agar.The value of pH was adjusted to 5.7-5.8.Effect of culture with Meristem tips from thermo-treated bulbs and the tips size on virus elimination are discussed.Cytogenetic analysis on vitro showed that there were 16 Chromosomes in all single cells observed,no variation was found. So the genetic type of material was maintained well.
