RAPD鉴定番茄一代杂种遗传纯度的研究

摘要:
以番茄优良一代杂交种毛粉808,毛粉818和强选一号为材料,提取DNA进行APD分析,在300个RAPD随机引物中筛选了可鉴定3个品种一代杂种遗传纯度的特异性引物,其中S1019,S1388和S1422可用于这3个品种的纯度鉴定,S1072可用于毛粉808和毛粉818的纯度鉴定,S1388可用于区分3个品种的父本和子代。

Identification of genetic purity for tomato F1 hybrids by means of RAPD
Abstract:
Extract DNA from three commerical tomato F1 hybrid cultivars,“Wool-Pink808”,“Wool-Pink818”,“Qiangxuanl” and their parents,these DNAs were subjected to RAPD screening with 300 RAPD primers,and DNA from other cultivars “Wool-Pink802” and “West-Pink3” were also used to RAPD analysis.Four primers S1019.S1072.S1388 and S1422 were found to be useful for identifying these cultivars's purity.Three of them could be used to identify the three commerical tomato F1 hybrid cultivars,s purity while primer S1072 could be used to indentify the purity of “Wool-Pink808” and “Wool-Pink818”.The primers S1388 could separate the male parents from the F1 hybrid of the three cultivars. Using RAPD marker will be the alternative method for phenotypic tests method to control tomato F1 hybrid's purity and to identify tomato cultivars.
