ABA和PEG对胡萝卜体细胞胚诱导和调控的影响

摘要:
对ABA和PEG诱导胡萝卜(Daucuscarota L.)体细胞胚发生及其调控体细胞胚发育的效应进行了研究。在MS液体培养基中(含蔗糖30g/L)附加10μmol/LABA或100g/LPEG6000诱导体细胞胚,能使其提早发生7~10 d,而且能使子叶胚处于调控状态。确定了ABA和PEG在MS液体培养基中(含蔗糖10 g/L)调控胡萝卜体细胞胚发育的下限浓度分别为1 μmol/LABA,200 g/L PEG6000,细胞学观察表明此种状态的胚缺乏营养。采用双向电泳分离出与ABA和PEG渗透调节密切相关的特异蛋白质多肽S1,分子质量27.9 ku,等电点6.0左右。

Effects of ABA and PEG on the embryogenesis and regulation of carrot (Daucus carota L.) somatic embryos
Abstract:
ABA and PEG effects on embryogenesis and regulation of carrot somatic embryos have been studied.When callus are suspended in MS liquid medium(containing 30 g/L sucrose and 10 μmol/L ABA or 100 g/L PEG 6000),the embryos can be promoted precociously by 7-10 days and the mature embryos be prevented from germination.Culturing in MS medium(containing 10 g/L sucrose),the regulated minimum concentrations of ABA and PEG 6000 are 1 μmol/L and 200 g/L respectively.The somatic embryo is short of nutration by cytology observation.Special proten S1,closely related to the osmotic regulation of ABA and PEG,is seperated (MW 27.9 ku,PI6.0).From these results,we have found the bases for researching about molecular mechanism of embryogensis in vitro and gene expression or regulation.
