大赖草和新麦草物种专化DNA重复序列研究Ⅰ. 分离和鉴定

摘要:
从本研究建立的Leymus racemosus ( Lam. ) Tzvelev(大赖草)和Psathyrostachys juncea ( Fisch) Nevski (新麦草)的DNA文库中分别挑出550和348个重组克隆提取质粒DNA,用普通小麦、 L.racemosusPsa.juncea等物种的总DNA作探针,通过两轮点渍杂交,发现有15个克隆稳定地与L.racemosus和(或)Psa.juncea有强杂交信号,与中国春无或基本无杂交信号,初步认定这些克隆为物种专化DNA重复序列克隆。其中5个又被选取作交互杂交以测定彼此间的同源性,结果表明,pPj605与pPj205有同源性,而pLr344,pLr426和pLr647相互间以及与pPj205和pPj605之间均无序列同源性。

A Study on Species-Specific DNA Repeats from Leymus racemosus and Psathyrostachys junceaⅠIsolation and Identification
Abstract:
550 and 348 recombinant clones were picked out from L.racemosus and Psa.juncea libraries and plasmid DNA was isolated from each of them.The dot blots of recombinant plasmid DNA were hybridized with the 32P labelled total DNA of Triticum aestivum,L.racemosus,Psa.juncea and other 4 species of Triticea.15 recombinant plasmids gave strong hybridization signals with DNA of L.racemosus and Psa.juncea but no or almost no hybridization signals with that of T.aestivum,which indicated that they could contain species-specific DNA repeats.Five of them were chosen for further characterization.pLr344,pLr426 and pLr647 were cloned from L.racemosus while pPj205 and pPj605 from Psa.juncea.Crosshybridization of the 5 DNA repeats indicated that pPj605 showed homology to pPj205.However,pLr344,pLr426 and pLr647 did not show homology,if any,to others.
