大白菜和油菜真空渗入遗传转化法初报

摘要:
在携带CaMV Cabb B-JI株系基因Ⅵ的根癌农杆菌菌种GV3101的介导下,对大白菜、油菜幼苗生理状态、真空渗入时间及不同处理部位和叶面造伤等的处理与遗传转化效果的关系进行了比较研究。结果表明,经自然春化后,大白菜、白菜型油菜植株长至4~5片叶,甘蓝型油菜、芥菜型油菜植株长至7~9片叶,花薹已抽出时,为真空渗入最适苗态。大白菜自然春化植株移栽缓苗(3 d),或者黑暗处理(8 h)均可显著提高其转化率。在吸力为14. 4m3/h的真空泵下处理1min后,迅速打开真空泵气阀升压变常压,再关闭阀门抽真空30 s的转化效果较好。仅处理叶片转化率可高达11. 20%,叶面造伤处理对转化率没有明显影响。

Vacuum Infiltration Genetic Transformation Method in Chinese Cabbage and Rape
Abstract:
The effects of the physiological state of seedling, vacuum infiltration time, parts of vacuum infiltration and artificial hurt on transformation were studied with the GV3101 strain of Agrobacterium tumefaciens carrying CaMV Cabb B-JIⅥin Chinese cabbage (B.campestris spp.pekinesis) and rape. It showed that it was the most suitable physiological state when the bolt had appeared and the naturally vernalized seedlings had 4-5 leaves in Chinese cabbage and B.campestris, and 7-9 leaves in B.napus and B.juncea. Both recuperating from the shock of transplanting and putting seedlings in darkness could increase the transformation frequence of naturally vernalized Chinese cabbage seedling. The transformation frequence was highest when drawing a vacuum for 1min with the sauction capacity of the vacuum pump was 14. 4m3/h.Only with the leaves treated the frequence of transformants could reach 11. 20%.Then releasing vacuum very rapidly, and closing the pump value to draw a vacuuam for another 30 seconds in B.campestris.Only the leaves treated could reach transformation frequence at 11. 20%.Artificial hurt had no effect on the transformation frequence.
