山羊小腔卵泡卵母细胞的体外成熟和体外受精

摘要:
采集繁殖和非繁殖季节山羊卵巢,回收大腔(3-6mm)和小腔(<2mm)卵泡卵母细胞,进行体外成熟和受精,结果如下:①繁殖季节的大腔和小腔卵泡卵母细胞体外成熟率(68.4%.,27.5%)均显著(P<0.05)高于非繁殖季节(52.5%,20.6%);②1和1.5周岁龄羊与成年羊相比,前两者的小腔卵泡卵母细胞采集数显著(P<0.05)多于后者(25.6,22.5:15.1),但卵母细胞体外成熟率无显著差异(P<0.05);③随卵母细胞直径增加,其体外成熟率呈显著上升趋势;④与加新生犊牛血清和生殖激素在颗粒细胞悬浮液中培养相比,加发情山羊血清和激素在颗粒细胞单层上培养可显著(P<0.05)提高小腔卵泡卵母细胞体外成熟率(20.8%,34.5%);⑤将体外成熟的小腔卵泡卵母细胞与体外获能精子进行受精,其受精率为35.7%(51/143)。

Maturation and Fertilization in Vitro of Goat Oocytes Derived from Small Antral Follicles
Abstract:
Goat ovaries were collected at breeding and nonbreeding seasons.The oocytes derived from large (LAF,3一6mm) and small antral follicles(SAF,<2mm) were recovered and used for maturation and fertilization in vitro.The results are as follows;① in vitro maturation rate of LAF and SAF oocytes is significantly higher at breeding seasons (68.4%,27.5%) than at nonbreeding seasons (52.5%,20.6%);② the numbers of SAF oocytes colleeted from the ovaries of goats at l (25.6) and 1.5 (22.5) years old is more than that from adult goat ovaries (15.1),but there is no significant difference in oocyte maturation rates among three groups;③in vitro maturation rates of oocytes increases significantaly with their diameter increase;④more high maturation rates of SAF oocytes is obtained when culture in medium with GS,FSH,LH and 17β一E2 on granulosa cell monolayer (34.5%) in comparison with culture in granulosa cell solotion with NCS,FSH,LH and 17β一E2 (20.8%);⑤SAF oocytes maturated were fertilization in vitro with capacited sperm and in vitro fertilization rate is 35.7% (51/143).
