褐藻寡糖对LPS诱导家兔肺脏损伤的保护作用

摘要:
【 目的】探究褐藻寡糖(AOS)对脂多糖(LPS)诱导家兔肺脏损伤的保护作用。【 方法】选用40只35日龄健康家兔,随机分为4组(每组10只):对照组、LPS组、AOS低剂量(100 mg/kg AOS)组和AOS高剂量(200 mg/kg AOS)组。预试期7 d,正试期 28 d。正试期第25-28天,LPS组及AOS组腹腔注射0.25 mg/(kg·d) LPS,对照组注射等体积PBS。试验结束,每组随机处死6只家兔,采集肺组织备用。采用HE染色观察肺脏组织形态学变化;采用ELISA试剂盒测定肺脏组织中TNF-α、IL-1β、IL-6、MDA和SP-C含量以及GSH-Px、SOD活性;免疫荧光检测肺泡上皮细胞SP-C蛋白表达水平。【 结果】HE染色显示,LPS组家兔肺脏组织出现出血、炎性细胞浸润及蛋白沉积;AOS低剂量和高剂量组家兔肺脏损伤均减轻。LPS组家兔肺脏组织中TNF-α、IL-1β、IL-6含量较对照组显著升高(P<0.05),2个AOS组上述因子含量较LPS组显著降低(P<0.05)。LPS组家兔肺脏MDA含量较对照组显著升高,GSH-Px及SOD活性显著降低(P<0.05);AOS低剂量和高剂量组家兔肺脏MDA较LPS组显著降低(P<0.05),GSH-Px和SOD活性显著升高(P<0.05),且AOS高剂量组SOD显著高于AOS低剂量组(P<0.05)。ELISA检测结果显示,LPS组家兔肺脏SP-C蛋白质量浓度较对照组显著降低(P<0.05);AOS低剂量和高剂量组家兔肺脏SP-C蛋白质量浓度较LPS组分别显著升高了21.59%和25.10%(P<0.05)。免疫荧光检测结果显示,LPS组家兔肺脏SP-C蛋白表达水平较对照组显著降低(P<0.05),AOS低剂量组SP-C蛋白表达水平较LPS组显著上调(P<0.05),且低剂量组高于高剂量组(P<0.05)。【 结论】饲料添加AOS可显著缓解LPS诱导的家兔肺脏损伤,保护肺脏功能。

Protective effects of alginate oligosaccharides on LPS⁃induced lung injury in rabbits

Abstract:
【Objective】This study aims to investigate the protective effects of alginate oligosaccharides (AOS) on lipopolysaccharide(LPS)-induced lung injury in rabbits.【Method】Forty healthy 35-day-old rabbits were randomly divided into four groups with 10 rabbits in each group:a control group,an LPS group,a low-dose AOS group(100 mg/kg AOS),and a high-dose AOS group(200 mg/kg AOS).The pretrial period lasted 7 days,followed by a 28-day experimental period. From days 25 to 28 of the experimental period,rabbits in the LPS and AOS groups were intraperitoneally injected with LPS at 0. 25 mg/(kg·d),while those in the control group received an equal volume of phosphate-buffered saline( PBS).At the end of the experiment,six rabbits from each group were randomly euthanized,and their lung tissues were collected for subsequent analysis.Histomorphological changes in lung tissues were observed using hematoxylin and eosin(HE) staining. The con-tents of tumor necrosis factor- α(TNF- α),interleukin-1β(IL-1β),interleukin-6(IL-6),malondialdehyde (MDA), and surfactant protein C(SP-C),as well as the activities of glutathione peroxidase(GSH-Px) and su?peroxide dismutase(SOD),were measured using enzyme-linked immunosorbent assay(ELISA) kits. The ex?pression of SP-C in alveolar epithelial cells was detected by immunofluorescence.【Result】HE staining revealed that lung tissues in the LPS group exhibited hemorrhage,inflammatory cell infiltration,and protein deposition,whereas these pathological changes were markedly alleviated in both the low-and high-dose AOS groups. Com?pared with the control group,the contents of TNF-α,IL-1β,and IL-6 in lung tissues were significantly increased in the LPS group(P<0.05),while these indicators were significantly decreased in the two AOS groups(P<0.05).Compared with the control group,the LPS group exhibited significantly higher MDA content and lower activities of GSH-Px and SOD(P<0. 05).Compared with the LPS group,both the low-and high-dose AOS groups showed significantly decreased MDA contents and increased GSH-Px and SOD activities(P<0.05),with the high-dose AOS group exhibiting significantly higher SOD activity than that in the low-dose AOS group (P<0.05).ELISA results demonstrated that the concentration of SP-C was significantly lower in the LPS group than in the control group(P<0.05),but was markedly increased by 21. 59% and 25. 10% in the low-dose and high-dose AOS groups,respectively(P<0. 05).Immunofluorescence analysis showed that SP-C pro-tein expression level was significantly lower in the LPS group than in the control group(P<0.05).Compared with the LPS group,both the low-and high-dose AOS groups showed markedly increased SP-C expression(P<0.05),with the increase being higher in the low-dose AOS group than that in the high-dose AOS group(P<0.05).【Conclusion】Dietary supplementation with AOS significantly alleviates LPS-induced lung injury and protects pulmonary function in rabbits.
