花楸树(Sorbus pohuashanensis)与北京花楸(S.discolor)远缘杂交亲和性解析

摘要:
【 目的】探究花楸树与北京花楸杂交亲和性,为花楸属植物杂交育种提供理论依据。【 方法】以花楸树 与北京花楸为材料,通过观测二者开花物候期、花部特征、人工杂交授粉试验以及荧光显微镜观察花粉管生长情况,研 究二者交配系统和正反交亲和性,并利用11对SSR标记引物鉴定杂交子代真实性。【 结果】花楸树的盛花期和末花 期晚于北京花楸,二者花期于4月中旬重合;花楸树与北京花楸平均胚珠数分别为6.70和9.27,平均单花花粉数分别为26 666.67和5 757.14粒,花粉胚珠比分别为3 980.10和621. 05,异交指数均为4。花楸树与北京花楸在开花前1 d和开花第1天柱头可授性较强。花楸树与北京花楸花粉均能在彼此柱头表面附着并萌发。花楸树×北京花楸组合比 北京花楸×花楸树组合的杂交亲和性更高。人工控制授粉试验结果表明,花楸树与北京花楸交配系统均为异交且自交不亲和,花楸树×北京花楸正反交杂交组合的平均结实率分别为44.10%和49.04%。SSR标记鉴定花楸树与北京 花楸正反交114个子代真实率为100%,仅需3对引物组合(sorsb3、sorse10、sorsd39)即可鉴定所有杂交子代。【 结论】花楸树与北京花楸交配系统为异交且自交不亲和,花楸树与北京花楸之间杂交亲和。

Analysis of distant cross⁃compatibility between Sorbus pohuashanensis and S.discolor

Abstract:
【 Objective】The study aims to investigate the cross-compatibility between Sorbus pohuashanensis and S.discolor,to provide a theoretical basis for hybrid breeding of Sorbus.【Method】The mating system and reciprocal cross-compatibility between S.pohuashanensis and S.discolor were investigated through observing flowering phenology,floral characteristics,artificial pollination experiments,and fluorescence microscope observation of pollen tube growth.The authenticity of hybrid progenies was identified using 11 pairs of SSR primers.【Result】The full-bloom stage and end of flowering period of S.pohuashanensis occurred later than those of S.discolor. with flowering periods overlapping in mid-April. The average ovule numbers were 6. 70 for S. pohuashanensis and 9.27 for S.discolor,while average pollen grains per floret were 26 666.67 and 5 757.14,respectively. The pollen-ovule ratio was 3 980.10 and 621.05,respectively,and the outcrossing index was 4 for both species. The stigmas of both species showed high receptivity one day before flowering and on the first flowering day. Pollen of both species was able to adhere to and germinate on reciprocal stigmas. Crossing combination of S.pohuashanensis×S.discolor exhibited higher compatibility than crossing combination of S.discolor×S.pohuashanensis.The artificial pollination experiment indicated that the mating systems of both species were outcrossing and self-incompatible,with average fruit-setting rates of 44.10%(S.pohuashanensis×S. discolor) and 49.04%(S.discolor×S. pohuashanensis).SSR marker analysis identified 114 hybrid progenies of S. pohuashanensis and S.discolor with 100% authenticity. Only three pairs of primer combinations(sorsb3,sorse10,sorsd39) were required to identify all the hybrid progenies.【Conclusion】Both S.pohuashanensis and S.discolor exhibited outcrossing and self-incompatible mating systems,and were reciprocally cross-compatible.
