| 天蚕素酵母菌体微胶囊制备及其在肠炎防治中的应用 |

| 【目的】开发天蚕素(cecropin)酵母菌体微胶囊(yeast-cell microcapsules,YCM),用于肠炎的防治,为“减抗”和“替抗”提供新策略。【方法】以酿酒酵母为载体,构建可表达天蚕素的真核表达载体,并通过醋酸锂转化法制备天蚕素YCM。利用荧光显微镜观察天蚕素-EGFP融合蛋白的表达;通过体外抑菌试验、胃肠液耐受性试验、肠道滞留时间试验检测天蚕素YCM的体外抑菌活性、对胃肠液耐受性及在肠道中的滞留时间。构建结肠炎小鼠模型,饲喂天蚕素YCM 7 d后评估天蚕素YCM对结肠长度和组织结构的影响,并通过16S rDNA测序分析天蚕素YCM对肠道菌群的影响。【结果】成功构建了8种天蚕素真核表达载体,并制备了具有分泌天蚕素能力的YCM。体外试验表明,天蚕素YCM对常见肠道致病菌具有抑菌活性,且在模拟胃肠液中表现出良好的耐受性。体内试验显示,天蚕素YCM可在小鼠肠道中滞留至少14 d,并可显著改善结肠炎小鼠的肠黏膜损伤程度,增大结肠长度,维持肠道菌群稳态。【结论】研制了天蚕素酵母菌体微胶囊,其在防治肠炎方面具有明显效果。 |

| 【Objective】The study aims to develop yeast-cell microcapsules (YCM) based on cecropin for the prevention and treatment of enteritis,to provide new strategies for reduction and replacement of antibiotics.【Method】Using Saccharomyces cerevisiae as the carrier,eukaryotic expression vectors for cecropin secretion were constructed, and cecropin YCM was prepared by the lithium acetate transformation method.The expression of the cecropin-EGFP fusion protein was observed via fluorescence microscopy.The in vitro antibacterial activity,gastrointestinal fluid tolerance,and intestinal residence time of cecropin YCM were assessed through corresponding tests.A colitis mouse model was established,and after 7 days of cecropin YCM feeding,the effects of cecropin YCM on colon length and tissue structure were evaluated.The impact of cecropin YCM on the gut microbiota was analyzed through 16S rDNA sequencing.【Result】Eight types of eukaryotic expression vectors for cecropin were successfully constructed,and YCM capable of secreting cecropin was prepared.In vitro tests demonstrated that cecropin YCM exhibited significant antimicrobial activity against common intestinal pathogens and showed good tolerance in simulated gastrointestinal fluids.In vivo tests revealed that cecropin YCM could stay in the mouse intestine for at least 14 days,significantly reduce the degree of intestinal mucosal damage,increase colon length,and maintain gut microbiota homeostasis in colitis mice.【Conclusion】The cecropin-based yeast-cell microcapsules were developed in this study,which have significant effect on the prevention and treatment of enteritis. |
