吕梁市微鞘藻科(蓝藻)形态学及分子系统学研究

摘要:
【目的】通过丰富微鞘藻科的物种多样性,进一步扩充中国藻种资源库,为推动现代蓝藻分类系统的单系化发展奠定基础。【方法】从中国山西省吕梁市的土壤、河流以及石头表面分离出53株丝状蓝藻样本,利用光学显微镜进行形态学观察,基于16S rRNA基因序列使用贝叶斯法、最大似然法以及邻接法构建系统发育树,基于16S-23S rRNA基因间隔区预测藻株的二级结构。【结果】结合形态学及分子系统学方法,将其中6株藻株鉴定为微鞘藻科物种,分别为具鞘微鞘藻、南极微鞘藻(Microcoleus antarcticus Casamatta & Johansen)、阿氏浮丝藻、伪秋微鞘藻、Planktothrix rubescens((De Candolle ex Gomont) Anagnostidis & Komárek)和Planktothrix suspensa((Pringsheim) Anagnostidis & Komárek)。【结论】南极微鞘藻是中国报道的蓝藻新记录种,该种的发现丰富了中国蓝藻物种多样性,为准确鉴定蓝藻提供了理论依据,也为微鞘藻科的进一步重新修订和评估打下基础。

Abstract:
【Objective】The study enriches the species diversity of Microcoleaceae and further expands the Chinese algal resource database,providing a foundation for promoting the monophyletic development of the modern cyanobacterial classification systems.【Method】Fifty-three filamentous cyanobacteria were isolated from soil,rivers,and stone surfaces in Lüliang City of Shanxi Province,China.The morphology of the algae strains was observed using light microscope.Phylogenetic trees were constructed based on the 16S rRNA gene sequences using the Bayesian method,maximum likelihood method and neighbor-joining method.The secondary structure of the algae strains was predicted based on the spacer of the 16S-23S rRNA genes.【Result】Using morphological and molecular systematics methods,six algae strains were identified as Microcoleaceae,including Microcoleus vaginatus,M.antarcticus,Planktothrix agardhii,M.pseudautumnalis,P.rubescens,and P.suspensa.【Conclusion】M.antarcticus is a newly recorded species of cyanobacteria in China.This discovery enriches cyanobacteria diversity in China,providing a basis for accurate identification of cyanobacteria,as well as laying a foundation for further revision and evaluation of Microcoleaceae.
