根边缘细胞对荞麦根尖莠去津毒害的缓解效应

摘要:
【目的】探究根边缘细胞(RBC)对荞麦根尖莠去津毒害的缓解效应。【方法】以晋荞8号(JB)和品甜荞2号(P2) 2个荞麦品种为材料,采用根悬空培养的方法,研究不同浓度莠去津对荞麦根边缘细胞数量、存活率、凋亡率以及初生根抗氧化系统、根系活力的影响。【结果】莠去津对荞麦根边缘细胞和幼根生长有毒害作用,其中7.5 mL/L莠去津处理时,晋荞8号(JB)和品甜荞2号(P2)的RBC 数量较其对照分别显著降低18.6%和32.0%,而其凋亡率较对照分别升高251.5%和328.3%。莠去津对荞麦根产生氧化胁迫伤害,诱导荞麦幼根抗氧化酶POD、SOD和CAT 活性升高,活性氧和MDA积累,根膜透性增大,而根系活力下降。其中3.75 mL/L莠去津处理时,与移除RBC的根尖相比,晋荞8号(JB)和品甜荞2号(P2)2个荞麦品种保留RBC的根尖H2O2含量分别降低45.6%和70.5%,O-2·含量分别降低33.6%和29.0%,SOD活性分别降低4.6 %和4.1%,POD活性分别降低36.0%和54.8%,CAT活性分别降低17.4%和16.9%,相对电导率分别降低54.2%和33.7%,MDA含量分别降低36.2%和55.1%,根系活力分别提高40.5%和29.8%。【结论】在一定浓度莠去津胁迫下,荞麦根尖通过产生较多的RBC,并通过RBC分泌更多的黏胶层螯合莠去津,从而减轻莠去津对根尖的伤害。

Alleviation effect of border cells on atrazine toxicity in Fagopyrum esculentum root tip

Abstract:
【Objective】This research aims to explore the alleviation effect of buckwheat root border cells (RBC) on the atrazine toxicity of buckwheat root tip.【Method】With two buckwheat varieties,Jinqiao 8 (JB) and Pintianqiao 2 (P2) as the study material,the effects of different concentrations atrazine on the number,survival rate,apoptosis rate of RBCs,antioxidant system of primary root and root activity of buckwheat root were analyzed.【Result】Atrazine had toxic effects on the growth of root border cells and young roots of buckwheat.Under treatment of 7.5 mL/L atrazine,compared with CK,the RBC numbers of JB and P2 were significantly reduced by 18.6% and 32.0%,respectively,and RBC apoptosis was increased by 251.5% and 328.3%,respectively.Atrazine caused oxidative stress damage to buckwheat roots,inducing the increase of antioxidant enzymes POD,SOD and CAT activities,the accumulation of reactive oxygen species and MDA,the increase of root membrane permeability,and the decrease of root activity.Under treatment with 3.75 mL/L atrazine,the H2O2 contents of RBC-retained root tips of two buckwheat cultivars,JB and P2,were 45.6% and 70.5% lower than those of RBC removed root tips.The O-2· contents of the RBC-retained root tips were 33.6% and 29.0% lower than those of RBC-removed root tips.The SOD activity in RBC-retained root tips of JB and P2 was 4.6% and 4.1% lower than that of RBC-removed root tips.The POD activity of RBC-retained root tips was 36.0% and 54.8% lower than that of RBC-removed root tips.The CAT activity of RBC-retained root tips was 17.4% and 16.9% lower than that of RBC-removed root tips.The relative conductivity of the RBC-retained root tips of JB and P2 was 54.2% and 33.7% lower than that of RBC-removed root tips.The MDA contents of RBC-retained root tips were 36.2% and 55.1% lower than those of RBC-removed root tips.The root activity of RBC-retained root tips of JB and P2 was 40.5% and 29.8% higher than that of RBC-removed root tips.【Conclusion】Under the atrazine stress of a certain concentration,buckwheat root tips mitigate damage by producing a greater number of root border cells (RBCs),which secrete more mucilage to chelate atrazine,thereby alleviating its toxic effects on the root tips.
