球虫感染对家兔小肠、盲肠和圆小囊形态结构及功能的影响

摘要:
【目的】探究球虫感染对家兔小肠、盲肠、圆小囊形态结构、功能及炎性因子含量的影响。【方法】将20只未被球虫感染的35日龄家兔随机分为对照组和感染组,感染组家兔一次性灌胃0.5×104个/mL大型艾美耳球虫卵囊的生理盐水2 mL,对照组家兔灌胃同等剂量的生理盐水。感染后第8天,每组随机抽取6只家兔处死,采集小肠(十二指肠、空肠、回肠)、盲肠以及圆小囊组织,采用HE染色观察家兔小肠、盲肠和圆小囊的组织形态学的变化,采用ELISA试剂盒检测各组织中白介素1β(IL-1β)、白介素6(IL-6)、肿瘤坏死因子α(TNF-α)等炎性因子的含量。【结果】与对照组相比,感染组家兔小肠和盲肠肠绒毛结构不完整,绒毛变短,隐窝加深等,空肠、回肠和盲肠观察到有不同发育阶段的球虫寄生;圆小囊发生炎性细胞浸润,且黏膜层有大量球虫寄生。ELISA检测结果显示,感染组家兔小肠、盲肠和圆小囊组织中的炎性因子TNF-α、IL-1β以及IL 6的含量极显著高于对照组(P<0.01)。【结论】大型艾美耳球虫感染可导致家兔的小肠、盲肠和圆小囊产生形态结构损伤与炎症反应,破坏家兔肠道的黏膜结构,促进炎性因子表达。

Effects of coccidiosis infection on the morphological structure and function of small intestine,cecum and round cyst in rabbits

Abstract:
【Objective】This research aimed to investigate the effects of coccidiosis infection on the morphology,function and inflammatory factors of small intestine,cecum and round cyst in rabbits.【Method】Twenty rabbits at the age of 35 days were randomly divided into control group and infection group.Rabbits in infection group were given 2 mL of physiological saline containing 0.5×104 Eimeria magna oocysts by gavage once.Rabbits in control group were given physiological saline of the same dose by gavage.On the 8th day of the experiment,6 rabbits were randomly selected from each group and sacrificed.Small intestine (duodenum,jejunum,ileum),cecum and sacculus rotundus tissues were collected for later use.The histomorphological changes of small intestine,caecum and sacculus rotundus were observed by HE staining,and the contents of IL-1 β,IL-6 and TNF-α in small intestine,caecum and sacculus rotundus tissues were detected by ELISA kit.【Result】Compared with the control group, the villi of small intestine and cecum of rabbits in infection group were incomplete.The villi became shorter,crypts deepened,coccidia parasitized in jejunum,ileum and cecum at different development stages.The inflammatory cells infiltrated into sacculus rotundus, and a large number of coccidia parasitized in mucosal layer.ELISA results showed that the inflammatory factors TNF-α,IL-1β and IL-6 in small intestine,cecum and sacculus rotundus tissues of infected rabbits were significantly higher than those in control group (P<0.01).【Conclusion】E.magna infection can cause morphological structure damage and inflammatory reaction in small intestine,cecum and sacculus rotundus of rabbits,destroy mucosal structure of rabbit intestine,and promote the expression of inflammatory factors.
