水芹4CL基因家族鉴定及高温干旱胁迫下的表达分析

摘要:
【目的】了解水芹Oja4CLs基因家族成员的特性以及在高温、干旱胁迫下的表达情况,为水芹Oja4CLs基因家族成员的功能研究提供理论依据。【方法】以拟南芥4CL保守序列搜索溧阳白芹基因组数据库,并对水芹4CL家族成员编码蛋白的理化性质及家族成员结构域和保守基序进行分析;同时研究高温、干旱胁迫对水芹Oja4CLs基因表达的影响。【结果】 在溧阳白芹基因组数据库中共鉴定得到77个水芹4CL家族成员,命名为Oja4CL01~Oja4CL77,其氨基酸残基数在249~2 318,分子质量为27.61~257.47 kD;等电点在5.17~8.82,二级结构α-螺旋占比26.54%~46.78%,β转角占比4.40%~10.61%,延伸链占比15.16%~25.33%,无规则卷曲占比29.53%~46.72%。亚细胞定位结果显示,25个Oja4CLs蛋白定位于细胞质,19个Oja4CLs蛋白定位于叶绿体,15个Oja4CLs蛋白定位于细胞质膜,12个Oja4CLs蛋白定位于原生质体,3个Oja4CLs蛋白定位于胞外,2个Oja4CLs蛋白定位于内质网,1个Oja4CLs蛋白定位于细胞核。水芹Oja4CLs基因对高温、干旱呈现不同的响应,定植后7 d(幼苗期),Oja4CL01、Oja4CL15、Oja4CL39、Oja4CL59在高温、干旱胁迫下大量表达;定植后28 d(成株期),Oja4CL59Oja4CL70在高温、干旱胁迫下大量表达;定植后56 d(开花期),Oja4CL15Oja4CL59在高温、干旱胁迫下大量表达。【结论】77个水芹4CL基因家族成员分为6个亚家族,大部分Oja4CLs都呈现较高表达,幼苗期水芹对高温、干旱比较敏感,高表达4CL基因以响应高温干旱。

Identification of 4CL gene family of Oenanthe javanica and its expression analysis under high temperature and drought

Abstract:
【Objective】This research aimed to understand the characteristics of Oja4CLs gene family members and their expression under high temperature and drought stress,to provide a theoretical basis for the functional study of the Oja4CLs gene family members.【Method】4CL conserved sequences of Arabidopsis were used to search the genome database of Liyang Oenanthe javanica,and the physical and chemical properties,domains and conserved motifs of the 4CL family members were analyzed.Effects of high temperature and drought stress on Oja4CLs gene expression were studied.【Result】A total of 77 4CL family members were identified in Liyang Oenanthe javanica genome database,named as Oja4CL01-Oja4CL77,whose number of amino acid residues ranged from 249-2 318,with molecular weight of 27.61-257.47 kD.The isoelectric point of protein ranged from 5.17-8.82, the secondary structure α -helix accounted for 26.54%-46.78%,the β angle accounted for 4.40%-10.61%,the extended chain accounted for 15.16%-25.33%,and the random coil accounted for 29.53%-46.72%.Subcellular localization results showed that 25 Oja4CLs proteins were located in the cytoplasm,19 Oja4CLs proteins were located in chloroplasts,15 Oja4CLs proteins were located in the plasma membrane,12 Oja4CLs proteins were located in protoplasts,3 Oja4CLs proteins were located in the extracellular,2 Oja4CLs proteins were located in the endoplasmic reticulum,and 1 Oja4CLs protein was located in the nucleus.Oja4CLs gene showed different responses to high temperature and drought.Oja4CL01,Oja4CL15,Oja4CL39 and Oja4CL59 were highly expressed at day 7 after planting (in seedling stage) under high temperature and drought stress.Oja4CL59 and Oja4CL70 were highly expressed at day 28 after implantation (in adult stage).Oja4CL15 and Oja4CL59 were highly expressed under high temperature and drought stress at day 56 after planting (in flowering stage).【Conclusion】77 members of 4CL gene family were divided into 6 subfamilies,and most of Oja4CLs were highly expressed.Oenanthe javanica was more sensitive to high temperature and drought in seedling stage,and the 4CL gene was highly expressed in response to high temperature and drought.
