1-丙硫醇对兰州百合采后篮状菌的防治效果

摘要:
【目的】探究寡养单胞菌KJ-6产生的挥发性有机化合物单体1-丙硫醇(1-Propanethiol)对兰州百合采后病原真菌篮状菌(Talaromyces tumuli)F-3的抑菌活性、抑菌机理及体内防治效果,为兰州百合鳞茎保鲜提供参考。【方法】以寡养单胞菌KJ-6产生的挥发性有机化合物单体1-丙硫醇为材料,以兰州百合采后病原真菌篮状菌(T.tumuli)F-3为靶标,采用熏蒸法测定不同浓度1-丙硫醇对篮状菌F-3的抑菌活性,采用扫描电镜(scanning electron microscopy,SEM)观察1-丙硫醇对篮状菌F-3菌丝形态的影响,采用碘化丙啶(propidium iodide,PI)染色法检测1-丙硫醇对篮状菌F-3细胞膜完整性的影响,采用紫外分光光度计测定不同浓度1-丙硫醇处理不同时间后篮状菌F-3在595和260 nm处的吸光度值,明确1-丙硫醇对篮状菌F-3蛋白和核酸泄露的影响。采用熏蒸法研究不同浓度1-丙硫醇对篮状菌F-3的体内防治效果,筛选1-丙硫醇对兰州百合的最佳熏蒸浓度。【结果】随着熏蒸浓度的升高,1-丙硫醇对篮状菌F 3菌丝生长的抑制率显著增大,当1-丙硫醇浓度为200 μL/L时,其对篮状菌F-3菌丝生长的抑制率高达88.51%。扫描电镜观察结果显示,1-丙硫醇处理导致篮状菌F-3菌丝发生明显塌陷、皱缩、扭曲折叠、内容物流出,菌丝失去线性。PI染色结果显示,经1-丙硫醇熏蒸的篮状菌F-3菌丝显示大量红色荧光,表明细胞膜完整性遭到严重破坏。随着篮状菌F-3在1-丙硫醇中暴露时间的延长及1-丙硫醇浓度的增加,篮状菌F 3菌丝蛋白和核酸的释放增加。1-丙硫醇对兰州百合采后病害的防治效果并没有完全随着熏蒸浓度的升高而增加,当熏蒸浓度为120 μL/L时,兰州百合病情指数最低,病害防治效果最好,防效高达86.77%。【结论】寡养单胞菌KJ-6挥发性有机化合物单体1-丙硫醇对兰州百合采后篮状菌F-3具有较好的抑制作用,1-丙硫醇通过改变篮状菌F-3的菌丝形态及破坏细胞膜完整性,引起菌丝体内容物外流,从而抑制病原真菌的生长。120 μL/L 1-丙硫醇熏蒸可以有效缓解兰州百合采后病害的发生,延长兰州百合货架期。

Control effect of 1-Propanethiol on Talaromyces tumuli in postharvest Lanzhou lily

Abstract:
【Objective】This research aimed to explore the antifungal activity,antifungal mechanism and in vivo control effect of 1-Propanethiol,the volatile organic compound monomer produced by Stenotrophomonas geniculata KJ-6 against pathogenic fungus Talaromyces tumuli F-3 in postharvest Lanzhou lily,to provide reference for keeping Lanzhou lily bulbs fresh.【Method】The volatile monomer 1-Propanethiol was used as the experimental material,and the pathogenic fungus T.tumuli F-3 in postharvest Lanzhou lily was used as the target fungus.The fumigation method was used to determine the antifungal activity of different concentrations of 1-Propanethiol against T.tumuli F-3.Scanning electron microscopy (SEM) was used to observe the effect of 1-Propanethiol on the mycelial morphology of T.tumuli F-3.Propidium iodide (PI) staining was used to detect the effect of 1-Propanethiol on T.tumuli F-3 cell membrane integrity.An ultraviolet spectrophotometer was used to measure the absorbance values of T.tumuli F-3 at 595 and 260 nm after the T.tumuli F-3 samples were treated with different concentrations of 1-Propanethiol for different time length,to clarify the effect of 1-Propanethiol on the leakage of T.tumuli F-3 protein and nucleic acid,and to analyze the antifungal mechanism of 1-Propanethiol.The fumigation method was used to study the in vivo control effects of different concentrations of 1-Propanethiol on T.tumuli F-3,to determine the optimal fumigation concentration of 1-Propanethiol for Lanzhou lily.【Result】The inhibition rate of 1-Propanethiol on mycelial growth of the pathogenic fungus T.tumuli F-3 increased significantly with the increase of fumigation concentration.At the fumigation concentration of 200 μL/L,the inhibition rate of mycelial growth reached 88.51%.SEM observation results showed that 1-Propanethiol treatment caused obvious collapse,shrinkage,twisting and folding of T.tumuli F-3 hyphae.The contents of T.tumuli F-3 flew out,and the hyphae lost linearity.The PI staining results showed that the hyphae of T.tumuli F-3 fumigated with 1 Propanethiol showed a large amount of red fluorescence,indicating that the integrity of the cell membrane was destroyed.As the exposure time of the pathogenic fungus T.tumuli F-3 to 1-Propanethiol was prolonged and the concentration of 1-Propanethiol increased,the release of mycelial protein and nucleic acid from T.tumuli F-3 increased.The control effect of 1-Propanethiol on postharvest diseases of Lanzhou lily did not always increase with the increase of fumigation concentration.When the fumigation concentration was 120 μL/L,the disease index of Lanzhou lily was the lowest and the disease control effect was the best,and the prevention effect is as high as 86.77%.【Conclusion】The volatile organic monomer 1-Propanethiol produced by S.geniculata KJ-6 showed a strong inhibitory effect on T.tumuli F-3 in postharvest Lanzhou lily.1-Propanethiol inhibited the growth of pathogenic fungus T.tumuli F-3 by altering the morphology of the mycelium and disrupting the integrity of the cell membrane,causing the outflow of mycelial contents.120 μL/L fumigation concentration of 1-Propanethiol can effectively alleviate the occurrence of postharvest diseases of Lanzhou lily and extend its shelf life.
