杜仲多糖对阿尔茨海默症小鼠脑组织神经炎症的改善作用

摘要:
【目的】研究杜仲多糖对阿尔茨海默症(Alzheimer’s diseas,AD)小鼠神经炎症的改善作用,为中药治疗阿尔茨海默症提供新思路。【方法】采用三相分离技术提取杜仲多糖。使用3月龄5×FAD小鼠作为AD模型小鼠,以3月龄C57BL/6J小鼠作为野生型小鼠。将小鼠分为野生型对照组、AD模型组和AD+杜仲多糖组,野生型对照组和AD模型组小鼠饲喂标准饲料,AD+杜仲多糖组小鼠饲喂添加0.25%(质量分数)杜仲多糖的标准饲料,小鼠饲喂至9月龄时终止。采集各组小鼠脑组织,使用免疫荧光染色对神经炎症相关小胶质细胞进行分析,探究杜仲多糖对AD小鼠神经炎症相关胶质细胞的改善作用。【结果】杜仲多糖可极显著抑制AD模型小鼠大脑海马区小胶质细胞和星形胶质细胞的异常增殖,改善小胶质细胞的形态,并可抑制海马区小胶质细胞的异常激活,逆转阿尔茨海默症小鼠中小胶质细胞形态向促炎表型的异常转变。【结论】杜仲多糖能够显著改善AD小鼠神经炎症相关胶质细胞的状态,具有开发为AD治疗药物的潜力。

Ameliorative effect of Eucommia cortex polysaccharides on neuroinflammation in the brain of Alzheimer’s disease mice

Abstract:
【Objective】This research aimed to study the ameliorative effect of Eucommiae cortex polysaccharides(EPs) on neuroinflammation-associated glial cells in Alzheimer’s disease mice,and to provide a new idea for the treatment of Alzheimer’s disease by traditional Chinese medicine.【Method】The three-phase separation technique was used to extract EPs.The 3-month-old 5×FAD mice were used as Alzheimer’s disease model mice,and the 3-month-old C57BL/6J mice were used as wild type mice.The mice were divided into 3 groups:wild-type control group,AD model group,and AD+EPs group.Mice in the AD+EPs group were fed standard chow supplemented with 0.25% EPs,and mice in the wild-type group and the AD group were fed standard chow until the mice were 9 months old.The brain of mice in each group were collected and analyzed with immunofluorescence staining for neuroinflammation-associated microglia to investigate the ameliorative effects of EPs on neuroinflammation-associated glial cells in mice with Alzheimer’s disease.【Result】EPs significantly inhibited the abnormal proliferation of microglia and astrocytes in the hippocampus of mice with Alzheimer’s disease,improved the morphology of microglia,and inhibited the abnormal activation of microglia in the hippocampus,which reversed the abnormal transformation of microglia morphology to a pro-inflammatory phenotype in mice with Alzheimer’s disease.【Conclusion】EPs significantly improves the status of neuroinflammation-associated glial cells in Alzheimer’s disease mice and is expected to be developed as a polysaccharide drug for Alzheimer’s disease treatment.
