【目的】基于非靶向代谢组学技术,对3个品种雪茄烟叶发酵前后的代谢物进行鉴定与分析,为雪茄烟叶发酵工艺研究提供理论依据。【方法】选择陕西汉中烟区川雪3号(CX3)、海研103(HY103)和青雪204(QX204)3个优质雪茄烟叶品种为试验材料,采用超高效液相色谱 四极杆飞行时间质谱联用技术(UHPLC-QTOF-MS),结合主成分分析(PCA)和正交偏最小二乘法 判别分析(OPLS DA)等多元统计分析方法,对不同品种雪茄烟叶发酵前后的代谢物进行代谢组学研究,筛选3个品种雪茄烟叶发酵前后的差异代谢物,并对筛选出的差异代谢物进行聚类分析、显著差异代谢物筛选和 KEGG 通路分析。【结果】CX3、HY103和QX204 3个品种雪茄烟叶发酵前后的代谢物相对含量存在显著差异,分别筛选出310,329和260种差异代谢物,主要为氨基酸、肽和类似物,有机酸,黄酮类化合物,脂质及生物碱。各品种中已鉴定的差异代谢物分别富集在74,74和65条KEGG通路中,主要的代谢通路为苯丙氨酸、酪氨酸和色氨酸生物合成,柠檬酸循环,抗坏血酸和醛糖代谢,吲哚生物碱生物合成,类黄酮生物合成,糖酵解/糖异生,甘油酯代谢,果糖和甘露糖代谢。【结论】3个品种雪茄烟叶发酵前后的代谢物存在一定差异,分别筛选鉴定了前20种差异代谢物,并获得了10条主要代谢通路。 |

【Objective】Based on non-targeted metabolomics approach,the metabolites in cigar leaves before and after fermentation were identified and analyzed to provide basis for improving fermentation process of cigar leaves.【Method】Three high quality cigar tobacco varieties of Chuanxue 3 (CX3),Haiyan 103 (HY103) and Qingxue 204 (QX204) were selected from the Hanzhong Tobacco District of Shaanxi.Ultra high performance liquid tandem chromatography quadrupole time of flight mass spectrometry (UHPLC-QTOF-MS) combined with principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA) and other multivariate statistical analysis was used for metabonomic analysis on leaves of three cigar varieties before and after fermentation.Differential metabolites in cigar leaves before and after fermentation were screened,and cluster analysis,significant difference metabolite screening and KEGG pathway analysis were performed.【Result】The contents of metabolites in CX3,HY103 and QX204 leaves before and after fermentation had significant differences,and 310,329 and 260 differential metabolites were screened,respectively.The metabolites were mainly classified into amino acids,peptides and analogues,organic acids,flavonoids,lipids and alkaloids.The identified differential metabolites were enriched in 74,74 and 65 KEGG pathways,respectively.Main metabolic pathways included phenylalanine,tyrosine and tryptophan biosynthesis,citrate cycle,ascorbate and aldarate metabolism,indole alkaloid biosynthesis,flavonoid biosynthesis,glycolysis/gluconeogenesis,glycerolipid metabolism,as well as fructose and mannose metabolism.【Conclusion】The metabolites of leaves of three cigar varieties before and after fermentation had certain differences.The first 20 differential metabolites were screened and identified,and 10 potential key metabolic pathways were obtained. |
