VC与外源激素浸种对滇油杉种子发芽及幼苗生长的影响

摘要:
【目的】研究不同质量浓度维生素C(vitamin C,VC)与外源激素赤霉素(GA3)和褪黑素(melatonin,MT)混合浸种不同时间对滇油杉(Keteleeria evelyniana)种子发芽及幼苗生长的影响,为滇油杉种子处理和壮苗培育提供科学依据。【方法】设置VC(A因素,1~4水平分别为0.0,1.5,2.0,2.5 g/L)、GA3(B因素,1~4水平分别为0.00,0.15,0.20,0.25 g/L)和MT(C因素,1~4水平分别为0.0,0.2,0.3,0.4 g/L)及其浸种时间(D因素,1~4水平分别为2,4,6,8 h)4个因素,采用U*12(1210)的变形U*12(410)均匀试验设计12个处理组合,以清水浸种8 h作为对照(CK),在种苗培育室内开展试验,分析不同质量浓度VC、GA3、MT混合溶液及浸种时间对滇油杉种子发芽性状、幼苗生长和根、干、叶及全株生物量的影响。【结果】13个处理组合的滇油杉种子发芽率、平均发芽时间和发芽指数分别为28.0%~44.0%、36.4~41.2 d和0.7~1.2粒/d;苗龄70 d时滇油杉幼苗的地径、苗高、胚轴长、针叶数、主根长和一级侧根数分别为0.41~0.59 mm、5.0~6.6 cm、3.2~4.1 cm、12.0~16.2针/株、8.4~13.1 cm和0.5~5.3条/株;根、干、针叶和全株生物量分别为0.008 2~0.013 6,0.005 2~0.008 1,0.013 1~0.026 0和0.028 4~0.047 6 g/株。不同处理组合间的以上指标均存在显著(P<0.05)或极显著(P<0.01)差异。VC是影响种子发芽性状的主导因子,幼苗生长和生物量主要受浸种时间的影响。0.0 g/L VC 、0.15 g/L GA3与0.3 g/L MT混合溶液浸种8 h(A1B2C3D4)能明显促进种子发芽,2.0 g/L VC、0.0 g/L GA3和0.2 g/L MT混合溶液浸种4 h(A3B1C2D2)能明显促进滇油杉幼苗生长和生物量积累。【结论】鉴于滇油杉苗期生长缓慢,其苗木培育以促进幼苗的生长为主要目的,因此处理组合A3B1C2D2可应用于生产实践中。

Effects of VC and exogenous hormone soaking on seed germination and seedling growth of Keteleeria evelyniana

Abstract:
【Objective】The effects of seed soaking with vitamin C (VC) and exogenous hormones of gibberellin (GA3) and melatonin (MT) for different times on seed germination and seedling growth of Keteleeria evelyniana were investigated to provide basis for seed treatment and sound seedling cultivation.【Method】 Four factors including VC (A,1-4 levels of 0.0,1.5,2.0 and 2.5 g/L),GA3 (B,1-4 levels of 0.00,0.15,0.20 and 0.25 g/L),MT (C,1-4 levels of 0.0,0.2,0.3 and 0.4 g/L) and soaking time (D,1-4 levels of 2,4,6 and 8 h) at 4 levels were used in the U*12(410) uniform experimental design,which was a variable form of the U*12(1210) design.Using water soaking seeds for 8 hours as the control (CK),the trial with 12 treatment combinations (TCs) was conducted in the laboratory to analyze the effects of different factors on germination characteristics,seedling growth and biomass amounts of root,stem,needle and whole seedling.【ResultThe germination percent (GP),mean germination time (MGT) and germination index (GIs) of 13 TCs were 28.0%-44.0%,36.4-41.2 days and 0.7-1.2 seed/day,respectively.Basal diameter (BD),seedling height (SH),hypocotyl length (HL),cotyledon number (CN),taproot length (TL) and primary lateral root number (PLRL) were 0.41-0.59 mm,5.0-6.6 cm,3.2-4.1 cm,12.0-16.2 needle/seedling,8.4-13.1 cm and 0.5-5.3 root/seedling,while biomass amounts of root,stem,needle and whole seedling of 70-day-old seedlings were 0.008 2-0.013 6,0.005 2-0.008 1,0.013 1-0.026 0 and 0.028 4-0.047 6 g/seedling,respectively.There were significant differences in above mentioned parameters (P<0.05 or P<0.01). VC was the dominant factor affecting seed germination traits,while growth and biomass of seedlings were primarily influenced by soaking time.Soaking seeds with mixed solution of 0.0 g/L VC 0.15 g/L GA3 and 0.3 g/L MT for 8 hours (A1B2C3D4) was beneficial for seed germination,while soaking with mixed solution of 2.0 g/L VC0.0 g/L GA3 and 0.2 g/L MT for 4 hours (A3B1C2D2) evidently promoted seedling growth and biomass accumulation.【Conclusion】Due to the slow growth of K. evelyniana seedlings,the seedling cultivation should focus on promoting seedling growth.Therefore,the A3B1C2D2 TC is suggested in production practice.
