| 【目的】筛选米槁(Cinnamomum migao)根部内生促生真菌菌株,探究其对米槁幼苗生长生理的影响。【方法】从贵州省黔南布依族苗族自治州的祥乐村、坝碰村和平艾村采集米槁鲜根,分离纯化得到内生真菌,利用透明圈法对所得菌株进行促生作用研究,将所得菌株接种至蒙金娜无机磷固体培养基、解钾固体培养基及King液体培养基进行促生能力测定,筛选促生能力强的菌株;再将筛选所得菌株分别回接至米槁幼苗,以接种等量空白培养基为对照,培养180 d后测定幼苗的生长生理指标。【结果】从米槁根部分离纯化得到41属177株内生真菌,利用透明圈法筛选得到16株具透明圈的真菌;促生能力测定结果表明,球毛壳菌(Chaetomium globosum)、哈茨木霉(Trichoderma harzianum)、深绿木霉(Trichoderma atroviride)及深色有隔内生真菌(Acrocalymma vagum)4株菌株具有较高促生潜力。接种试验结果表明,球毛壳菌能显著促进米槁幼苗对钾的吸收,哈茨木霉能显著促进过氧化氢酶(CAT)活性及幼苗对磷的吸收,深绿木霉能显著促进米槁幼苗苗高及地下部分的生长,深色有隔内生真菌能显著促进幼苗SOD活性。4种促生菌对米槁幼苗的促生作用不同,其中促生菌的产IAA能力与米槁幼苗的生长显著相关,溶磷、解钾能力与PPO活性呈显著负相关(P<0.05)。【结论】球毛壳菌、哈茨木霉、深绿木霉及深色有隔内生真菌对米槁幼苗生长均有促进作用,但不同菌种对米槁幼苗生长发育的作用机理不同。 |

| 【Objective】Endophytic growth-promoting fungus strains in roots of Cinnamomum migao were screened and their effects on seedling growth were investigated.【Method】Fresh roots of C. magao were collected from Xiangle village,Bapeng village and Ping’ai village in Qiannan Buyi and Miao Autonomous Prefecture of Guizhou Province,and endophytic fungi were isolated and purified.The growth promoting effect of selected strains was studied by transparent circle method.The obtained strains were inoculated into the solid medium of inorganic phosphorus, solid medium of potassium hydrolysis and King liquid medium for the determination of growth promotion ability.The strains with strong growth promotion ability were screened and grafted back to C. migao seedlings,using inoculation of blank medium as the control. After culturing for 180 days,the growth physiological indexes of C. migao seedlings were measured.【Result】A total of 177 endophytic fungi belonging to 41 genera were isolated and purified from C.migao roots,and 16 fungi with transparent rings were screened to study the growth promoting potentials.Chaetomium globosum,Trichoderma harzianum,Trichoderma atroviride and Acrocalymma vagum were found with strong growth promoting ability.C. globosum promoted potassium absorption seedlings and T. harzianum significantly promoted the activity of CAT enzyme and phosphorus absorption of seedlings.T. atroviride promoted the growth of seedling heights and underground parts and the dark septum endophytic fungus A. vagum promoted activities of SOD enzyme in seedlings.The four growth-promoting strains had different effects on seedlings. The IAA production capacity of the growth-promoting fungi was significantly correlated with seedling growth and the phospho-solubilizing and potassium releasing capacity was significantly negative correlated with PPO enzyme activity of seedlings (P<0.05).【Conclusion】The obtained four growth promoting strains of C. globosum,T. atroviride,T. harzianum and A. vagum all promoted the growth of C. migao seedlings with different impact levels. |
