药用真菌桑黄抗氧化物质及其活性研究

摘要:
【目的】探索杨树桑黄、鲍姆桑黄和桑树桑黄3个菌株的抗氧化活性成分和抗氧化能力,为药用真菌桑黄的充分利用提供依据。【方法】以桑黄孔菌属的杨树桑黄(Sanghuangporus vaninii)、鲍姆桑黄(S.baumii)和桑树桑黄(S.sanghuang)为研究对象,液体培养21 d,每隔3 d测定菌丝生长量及发酵液中多糖、多酚、黄酮和抗坏血酸含量,并分析发酵上清液的DPPH自由基、ABTS自由基、羟自由基和超氧阴离子清除能力以及铁离子还原能力、亚铁离子螯合能力、总抗氧化能力和超氧化物歧化酶(SOD)活性,分析抗氧化物质含量与抗氧化指标间的相关性。【结果】3个桑黄菌株发酵液均具有较强的抗氧化能力,但不同菌株的抗氧化活性成分含量和抗氧化指标的强弱存在很大差异。其中杨树桑黄的DPPH、ABTS自由基和超氧阴离子清除能力、亚铁离子螯合能力及SOD活性更强,鲍姆桑黄的铁离子还原能力更强,桑树桑黄的羟自由基清除能力、总抗氧化能力更强,杨树桑黄的抗氧化活性整体优于鲍姆桑黄和桑树桑黄。Pearson分析结果显示,多糖含量与DPPH自由基清除能力、铁离子还原能力、亚铁离子螯合能力呈极显著正相关,多酚含量与DPPH自由基清除能力、ABTS自由基清除能力、铁离子还原能力、超氧阴离子清除能力、亚铁离子螯合能力和SOD活性呈极显著正相关;黄酮含量与铁离子还原能力、羟自由基清除能力、超氧阴离子清除能力和总抗氧化能力呈极显著正相关;抗坏血酸含量与DPPH自由基清除能力、ABTS自由基清除能力、羟自由基清除能力、超氧阴离子清除能力和SOD活性呈极显著正相关。【结论】杨树桑黄、鲍姆桑黄和桑树桑黄均具有较强的抗氧化活性,且杨树桑黄表现更好。多酚、黄酮和抗坏血酸含量均可作为评价桑黄抗氧化活性的主要指标。

Antioxidant substances and activity of medicinal fungus Sanghuangporus

Abstract:
【Objective】This study explored the active components and antioxidant capacity of three strains including Sanghuangporus vaninii,Sanghuangporus baumii and Sanghuangporus sanghuang to provide basis for the protection and development of Sanghuangporus sanghuang germplasm resources.【Method】During the 21 days of liquid culture,the growth of mycelia and contents of polysaccharides,polyphenols,flavonoids and ascorbic acid in the fermentation supernatant of Sanghuangporus vaninii,S.baumii and S.sanghuang were measured every three days.The DPPH free radical scavenging ability,ABTS free radical scavenging ability, hydroxyl radical scavenging ability,superoxide anion scavenging ability,FRAP,ferrous ion chelating ability,total antioxidant capacity and SOD activity of the fermentation supernatants were determined.The relationships between contents of active components and antioxidant indexes were also analyzed.【Result】The fermentation broths of all three Sanghuangporus fungi had strong antioxidant capacity with large differences in active ingredients and antioxidant indexes among strains.S. vainii had a stronger ability to scavenge DPPH radical,ABTS radical and superoxide anions and to chelate ferrous ions with higher superoxide dismutase(SOD) activity.S.baumii had a stronger reduction ability of ferrous ion,while S.sanghuang had a stronger ability to scavenge hydroxyl radical and better capacity of total anti-oxidation.Pearson analysis showed that the accumulation of polysaccharides had significantly positive correlation with the ability of DPPH radical scavenging, iron reduction and ferrous chelating.The content of polyphenols had significantly positive correlation with the ability of DPPH radical,ABTS radical and superoxide anion scavenging, iron reduction ability,ferrous chelating ability as well as SOD activity.The content of flavonoids was positively correlated with the ability of iron reduction,hydroxyl radical and superoxide anion scavenging as well as total anti-oxidation.The content of ascorbic acid was positively correlated with the ability of DPPH radical,ABTS radical, hydroxyl radical scavenging as well as SOD activity.【Conclusion】Sanghuangporus vaninii,S.baumii and S.sanghuang had strong antioxidant capacity,and S.vaninii was better than S.baumii and S.sanghuang.Contents of polyphenols,flavonoids and ascorbic acid can be used as main evaluation indexes of antioxidant ability of Sanghuangporus.
