光照强度对菊花试管苗生长和抗氧化酶活性的影响

摘要:
【目的】研究不同光照强度的LED红蓝光质对菊花试管苗生长和抗氧化酶活性的影响,为高品质菊花组培苗的生产提供理论依据。【方法】以菊花试管苗为材料,在70%R+30%B的红蓝光质配比下,设置5种不同光照强度(15,30,45,60,75 μmol/(m2·s)),以普通荧光灯作为对照(CK),探究固定红蓝光质条件下不同光照强度对菊花试管苗形态指标、光合色素含量、根系活力、可溶性糖含量、可溶性蛋白含量以及抗氧化酶活性的影响。【结果】菊花试管苗生理生化指标在不同光照强度下表现不同。其中,叶数、叶长、叶幅及SOD活性均在75 μmol/(m2·s)处理下达到最大值,叶绿素a、叶绿素b、叶绿素总含量和类胡萝卜素含量以及可溶性蛋白含量均在30 μmol/(m2·s)处理下达到最大值,POD活性和MDA含量以60 μmol/(m2·s)处理最高,株高、根系活力和可溶性糖含量均在45 μmol/(m2·s)处理下达到最大值。【结论】在菊花试管苗生根阶段选用45 μmol/(m2·s)的光照强度,更利于其根系生长;而在菊花试管苗生长发育其他阶段,选用75 μmol/(m2·s)的光照强度更适宜其生长发育。

Effect of light intensity on growth and antioxidant enzyme activities of Chrysanthemum morifolium in vitro

Abstract:
【Objective】To provide basis for production of high quality Chrysanthemum morifolium plantlets in vitro,the effects of light intensity on growth and antioxidant enzyme activities of C. morifolium plantlets were studied under LED red and blue light conditions.【Method】Under the red and blue light with ratio of 70% R+30% B,the C. morifolium plantlets in vitro were cultured under five intensities of 15,30,45,60 and 75 μmol/(m2·s) with ordinary fluorescent lamp as CK.Then,the effects of different treatments on morphology,photosynthetic pigment content,root activity,soluble sugar content,soluble protein content and antioxidant enzyme activities of C. morifolium plantlets in vitro were explored.【Result】The physiological and biochemical indexes of C. morifolium plantlets in vitro behaved differently under different light intensities.Among them,the number of leaves,leaf length,leaf width and SOD activity of C. morifolium plantlets in vitro reached the maximum under the treatment of 75 μmol/(m2·s).The chlorophyll a, chlorophyll b,total chlorophyll content,carotenoid content and soluble protein content were highest under the treatment of 30 μmol/(m2·s).The highest POD activity and MDA content were under the treatment of 60 μmol/(m2·s),and plant height,root activity and soluble sugar content reached the maximum under the treatment of 45 μmol/(m2·s).【Conclusion】The light intensity of 45 μmol/(m2·s) was suggested at the rooting stage to improve root growth,while the intensity of 75 μmol/(m2·s) was recommended at other growth and development stages for growth and development.
