HIF-1α基因第10外显子对藏绵羊高原低氧适应性的影响

摘要:
【目的】研究藏绵羊低氧诱导因子1α基因(HIF-1α)第10外显子的变异特征及其对藏绵羊血气和生化指标的影响,为藏绵羊低氧适应性机制研究提供基础。【方法】采用PCR-SSCP法对333只藏绵羊HIF-1α基因第10外显子变异进行分析;采用血气仪测定其中134只年龄基本一致成年母羊的血气和生化指标,分析HIF-1α基因第10外显子变异与藏绵羊血气和生化指标的相关性。【结果】藏绵羊HIF-1α基因第10外显子存在6个SNPs位点,其中4个SNPs为同义突变,位于编码区(c.1302C>T、c.1392G>A、c.1506T>C和c.1515T>G),另外2个SNPs位于非编码区(c.1545+26A>G和c.1545+52G>A)。6个SNPs构成A、B、C 3个等位基因,表现为AA、BB、AB、AC和BC 5种基因型,其中AB为优势基因型。遗传多样性分析表明,HIF-1α基因第10外显子多态信息含量为0.48,呈中度多态,并处于Hardy Weinberg不平衡状态。HIF-1α基因第10外显子基因型对酸碱度(pH)、碱剩余(BE)、血氧饱和度(S(O2))、血红蛋白总量(Hb)和半饱和氧分压(p50)影响显著。AB基因型藏绵羊在群体中比例最高,且具有较高的S(O2)。与AB基因型藏绵羊相比,AA基因型藏绵羊BE和S(O2)较低(P<0.05),推测其高原低氧适应能力较弱。【结论】HIF-1α基因第10外显子变异与藏绵羊pH、BE、S(O2)、Hb和p50相关,推测AB基因型藏绵羊具有较好的高原低氧适应性,而AA基因型藏绵羊高原低氧适应能力较弱。

Effect of HIF-1α gene exon 10 on plateau hypoxia adaptability of Tibetan sheep

Abstract:
【Objective】The characteristics of HIF-1α gene exon 10 and its effects on blood gas biochemical indexes in Tibetan sheep were studied to provide basis for understanding hypoxic adaptation mechanism of Tibetan sheep.【Method】The variation of HIF-1α gene exon 10 in 333 Tibetan sheep was analyzed by PCR-SSCP.The blood gas biochemical indexes of 134 adult ewes at same age were measured by blood gas analyzer.The correlation between gene variation and blood gas biochemical indexes of Tibetan sheep was also analyzed.【Result】There were 6 SNPs in the HIF-1α gene exon 10 with four synonymous mutations located in coding region (c.1302C>T,c.1392G>A,c.1506T>C and c.1515T>G) and two located in non-coding region (c.1545+26A>G and c.1545+52G>A).These SNPs formed 3 alleles (A,B and C) with 5 genotypes (AA,BB,AB,AC and BC),and AB was preponderant genotype.Genetic diversity analysis showed that HIF-1α gene exon 10 was moderately polymorphic with the PIC of 0.48 and was in an imbalance of Hardy Weinberg.Correlation analysis showed that HIF-1α gene exon 10 mutation was associated with pH,base residual (BE),oxygen saturation (S(O2)),total hemoglobin (Hb) and partial saturation oxygen (p50).Sheep with genotype AB had the highest proportion in the Tibetan sheep population and high S(O2).Compared with the AB genotype sheep,BE and S(O2) of the AA genotype Tibetan sheep were lower (P<0.05) with possibly weak plateau hypoxia adaptability.【Conclusion】The variation of HIF-1α gene exon 10 was correlated with pH,BE,S(O2),Hb and p50 of Tibetan sheep.It was inferred that AB genotype Tibetan sheep had better plateau hypoxia adaptability,while AA genotype Tibetan sheep had weaker plateau hypoxia adaptability.
