山刺玫叶总黄酮的抗衰老作用研究

摘要:
【目的】研究山刺玫叶总黄酮的抗衰老作用机制,为山刺玫叶的开发利用提供参考。【方法】选取SPF级昆明种小鼠60只,雌雄各半,随机分为空白组、衰老模型组、维生素E组和山刺玫叶总黄酮低、中、高剂量组共6组(n=10),其中空白组小鼠用等体积生理盐水灌胃;衰老模型组小鼠颈背部皮下注射125 mg/(kg·d) D-半乳糖,灌胃等体积生理盐水;维生素E组小鼠颈背部皮下注射125 mg/(kg·d) D-半乳糖,灌胃维生素E溶液50 mg/(kg·d);山刺玫叶总黄酮低、中、高剂量组小鼠均颈背部皮下注射125 mg/(kg·d) D-半乳糖,并依次灌胃山刺玫叶总黄酮溶液1.0,2.0,4.0 g/(kg·d),同样条件下饲养。6周后分别测定各组小鼠免疫器官指数及血清、脑和肝脏组织中的SOD、GSH Px活性和MDA浓度,HE染色法观察各组小鼠脑组织的形态学变化,ELISA法检测各组小鼠血清中细胞因子IL-2、IL-4、IL-6、TNF-α的质量浓度。【结果】山刺玫叶总黄酮能显著或极显著提高衰老模型组小鼠脑、胸腺和脾脏指数(P<0.05,P<0.01),显著或极显著提高其脑、肝脏、血清中的SOD、GSH-Px活性(P<0.05,P<0.01),同时显著或极显著降低小鼠肝脏、血清和脑组织中的MDA浓度(P<0.05,P<0.01)。与衰老模型组比较,山刺玫叶总黄酮剂量组小鼠脑组织的神经元细胞数量增加,细胞的坏死及变性情况减轻,且随着山刺玫叶总黄酮剂量的增加恢复程度逐渐增加,即存在剂量依赖性。山刺玫叶总黄酮高、中剂量组小鼠血清中细胞因子IL-2、IL-4的质量浓度显著或极显著高于衰老模型组小鼠(P<0.05,P<0.01),而IL-6、TNF-α的质量浓度则显著或极显著低于衰老模型组小鼠(P<0.05,P<0.01)。【结论】山刺玫叶总黄酮对机体的衰老有延缓作用,其机制与清除过多自由基、预防脂质过氧化及增强免疫功能有关。

Anti aging effect of total flavonoids from leaves of Rosa davurica Pall.

Abstract:
【Objective】The anti-aging mechanism of total flavonoids from leaves of Rosa davurica Pall.was investigated to provide reference for development and utilization of Rosa davurica Pall.leaves.【Method】A total of 60 SPF Kunming mice with half male and half female were randomly divided into six groups,including blank group,aging model group,vitamin E group as well as low,medium and high dose groups with total flavonoids from leaves of Rosa davurica Pall.(n=10).The blank group mice were gavaged with equal volume of normal saline.The aging model group mice were subcutaneously injected D-galactose 125 mg/(kg·d) on back of neck and gavaged with equal volume of normal saline.The vitamin E group mice were treated same as aging group but gavaged with vitamin E (50 mg/(kg·d)).The total flavonoids groups had same injection of D-galactose and gavaged with total flavonoids contents of 1.0,2.0,and 4.0 g/(kg·d) successively.All mice were fed under same conditions.After 6 weeks,the immune organ index of each group mice was determined.Activities of SOD and GSH Px enzymes and MDA content in serum,brain and liver tissues were measured.Morphological changes of brain tissue were observed by HE staining and mass concentrations of cytokines IL-2,IL-4,IL-6 and TNF-α in serum were detected by ELISA.【Result】 The total flavonoids from leaves of Rosa davurica Pall.significantly or very significantly increased brain,thymus and spleen indexes of aging model group mice (P<0.05,P<0.01),and enzyme activities of SOD and GSH Px in brain,liver and serum were significantly or very sifnificantly increased (P<0.05,P<0.01),while decreased MDA content in tissues significantly (P<0.05,P<0.01).Compared with the aging model group mice,the number of neurons in brain tissue and the reduction of cell necrosis and degeneration of mice in total flavonoids groups were dose dependent.Serum cytokines IL-2 and IL-4 of high and middle dose groups mice were significantly or very significantly higher than that of the aging model group mice,while IL-6 and TNF-α were opposite (P<0.05,P<0.01).【Conclusion】The total flavonoids from leaves of Rosa davurica Pall.had effect of delaying organ senility and the mechanism was related with removing excess free radicals,preventing lipid peroxidation and enhancing immune function.
