大白菜菌核病抗性鉴定方法研究

摘要:
【目的】探讨鉴定大白菜菌核病抗病性的简易方法,为大白菜抗菌核病育种提供借鉴。【方法】以大白菜菌核病样病原菌为接种体,选用抗病品种15S261和15S94、耐病品种14S474和14S265及感病品种14S406、15S64和17S20为试验材料,对大白菜离体叶片和活体幼苗进行抗病性鉴定,并对离体叶片的接种时期、发病温度、接种部位及活体幼苗的接种时期、接种量进行比较,最后利用离体叶片接种法对89份大白菜种质资源进行菌核病抗病性鉴定。【结果】大白菜离体叶片抗病性鉴定的最适接种苗龄为4叶期,最适宜接种部位为叶片正面,最佳发病温度为25 ℃;活体幼苗抗病性鉴定的最适接种量为直径5 mm的菌丝琼脂块,最适接种苗龄为3叶期。在89份被鉴定的大白菜种质材料中,23份材料表现抗病,38份材料表现耐病,21份材料表现感病,7份材料表现高感。【结论】离体和活体菌丝琼脂块接种方法均能准确鉴定出大白菜的抗病性,其中离体菌丝琼脂块法更经济适用;大白菜种质资源中存在着抗病和耐病材料,可用于抗(耐)菌核病品种的选育。

Identification method of resistance to Sclerotinia in Chinese cabbage

Abstract:
【Objective】This study proposed a new method to detect resistance of Chinese cabbage to Sclerotinia to provide basis for disease resistance breeding of Chinese cabbage.【Method】Resistant varieties 15S261 and 15S94,moderately resistant varieties 14S474 and 14S265,and susceptible varieties 14S406,15S64 and 17S20 were used as inoculums to establish an artificial inoculation method for identification of resistance at seedling stage.The inoculation age,inoculation site and inoculation culture temperature,inoculation seedling and inoculation size of hyphae agar block for identification of seedling disease resistance of S.sclerotiorum were also compared.Then,the resistance of 89 Chinese cabbage lines were identified by this method.【Result】For identification of in vitro leaf disease resistance of S. sclerotiorum in Chinese cabbage,the optimal inoculation age was four-leaf-stage,the optimal inoculation site was front of leaf blade,and the optimal culture temperature was 25 ℃.For identification of seedling disease resistance,the optimal inoculation size of hyphae agar block was 5 mm in diameter and the optimal inoculation seedling was three-leaf-stage.A total of 23 resistance lines,38 tolerance lines,21 susceptible lines and 7 high susceptible lines were identified from 89 Chinese cabbage lines.【Conclusion】Both methods of disease resistance identification in vitro leaf and at seedling stage were effective in discrimination of resistance of Chinese cabbage germplasm.There are resistant and tolerant lines in Chinese cabbage,which can be used for breeding of resistant (tolerant) varieties.
