白花败酱草醇提物对四氯化碳所致小鼠急性肝损伤的保护作用

摘要:
【目的】研究白花败酱草醇提物对四氯化碳(CCl4)诱导小鼠急性肝损伤的保护作用,为白花败酱草醇提物的药用及作为保健野菜的开发利用提供参考。【方法】以40只洁净级昆明小鼠为研究对象,设空白对照组、模型组、白花败酱草醇提物低剂量组、中剂量组、高剂量组5个处理,用CCl4腹腔注射诱导建立小鼠急性肝损伤模型,采用苏木精 伊红(HE)染色法对肝脏组织进行病理检测;采用原位末端转移酶标记技术(TUNEL)染色检测小鼠肝细胞凋亡情况;采用蛋白质免疫印迹(Western blot)检测小鼠肝组织细胞凋亡相关蛋白的表达水平。【结果】白花败酱草醇提物能显著改善肝组织病变情况,显著降低小鼠血清中谷丙转氨酶(ALT)和天门冬氨酸氨基转移酶(AST)的活性及肝匀浆中丙二醛(MDA)含量和乳酸脱氢酶(LDH)活性,显著提高小鼠肝匀浆中超氧化物歧化酶(T-SOD)活性、总抗氧化能力(T-AOC)和谷胱甘肽(GSH)含量。TUNEL染色结果显示,白花败酱草醇提物能显著抑制CCl4诱导的肝细胞凋亡。与模型组相比,白花败酱草醇提物使CCl4刺激的小鼠肝组织Bcl-2蛋白表达显著升高,Bax蛋白表达显著降低,Bcl-2/Bax升高。【结论】白花败酱草醇提物通过抗氧化及抑制肝细胞凋亡而改善CCl4诱导的小鼠急性肝损伤。

Protective effects of alcohol extract of Patrinia villosa Juss on acute liver injury in mice induced by carbon tetrachloride

Abstract:
【Objective】This study evaluated the protective effects of Patrinia villosa Juss extract on carbon tetrachloride (CCl4)induced hepatic injury in mice.To provide reference for the medicinal use of alcohol extract of Patrinia villosa Juss and its development and utilization as a healthwild vegetable.【Method】40 clean kunming mice were divided into blank control group,model group,low-dose group,moderate-dose group and high-dose group of alcohol extract of Patrinia villosa Juss.Intraperitoneal injection of CCl4 was conducted to induce acute livery injury in a mouse model.Liver histopathology was confirmed using hematoxylin and eosin staining,and hepatocyte apoptosis was detected by TUNEL staining.Western blot analysis was used to determine the level of apoptosis related protein in liver tissues.【Result】Patrinia villosa Juss extract significantly improved the liver tissue lesions.It significantly reduced serum alanine aminotransferase and aspartate aminotransferase activity, malondialdehyde content,and lactate dehydrogenase activity in the liver homogenate,and it increased the total superoxide dismutase activity,total antioxidative capacity,and glutathione content.TUNEL staining showed that Patrinia villosa Juss extract significantly inhibited CCl4 induced hepatocyte apoptosis.Patrinia villosa Juss extract significantly elevated Bcl-2 protein expression and reduced the level of Bax protein expression in the livers of mice with CCl4 induced hepatic injury, leading to increase of Bcl-2/Bax.【Conclusion】Patrinia villosa Juss extract mitigated CCl4 induced acute hepatic injury in mice through antioxidation and inhibition of hepatocyte apoptosis.
