灰树花提取物的体内抗肿瘤及免疫活性研究

摘要:
【目的】研究灰树花不同极性溶剂提取物在小鼠体内的抗肿瘤和机体免疫活性。【方法】按极性从低到高依次采用石油醚、乙酸乙酯、丙酮和甲醇加热回流提取灰树花子实体,获得石油醚提取物、乙酸乙酯提取物、丙酮提取物和甲醇提取物。70只SPF级昆明小鼠(雌雄各半)分为7组,每组10只,分别标记为空白组、模型组、CTX阳性组及灰树花子实体石油醚、乙酸乙酯、丙酮及甲醇提取物组,小鼠接瘤后第2天开始给药,空白组不给药及生理盐水,模型组给予生理盐水,CTX阳性组隔天腹腔注射CTX 20 mg/kg,石油醚、乙酸乙酯、丙酮和甲醇提取物组每天灌胃相应的灰树花子实体提取物,抑瘤10 d。通过检测肿瘤抑制率,脏器指数,血清中影响因子干扰素-γ(INF-γ)、白细胞介素-2(IL-2)、血管内皮生长因子(VEGF)、过氧化氢酶(CAT)、丙二醛(MDA)和超氧化物歧化酶(SOD)水平等指标及肿瘤切片观察,研究灰树花子实体不同极性溶剂提取物对Hep-A-22荷瘤小鼠体内抗肿瘤、免疫活性及抗氧化作用。同时,通过小鼠耳肿胀试验和腹腔巨噬细胞吞噬试验进一步探究灰树花子实体提取物的免疫活性作用。【结果】各给药组均具有抑制肿瘤生长作用,其中灰树花子实体甲醇提取物组抑瘤效果最佳(52.31%),与模型组相比有极显著性差异(P<0.01),该组血清中IL-2、INF-γ和SOD含量极显著增加(P<0.01),CAT含量显著增加(P<0.05),并与抑制肿瘤具有相关性。通过HE染色和荧光染色后的肿瘤细胞观察,发现肿瘤细胞出现大面积坏死、消融等现象;各给药组对小鼠二甲苯耳肿胀均有抑制效果,其中甲醇组和乙酸乙酯组效果较好,与空白组差异性极显著(P<0.01)。【结论】灰树花甲醇提取物和乙酸乙酯提取物具有良好的抑制肿瘤生长效果。甲醇提取物组能够显著提高小鼠腹腔巨噬细胞活性,从而提高机体免疫活性,并与其抗肿瘤作用具有相关性。

Antitumor and immune activities of extracts from Grifola frondosa in vivo

Abstract:
【Objective】This study investigated the antitumor and immune activities of different polar solvent extracts from Grifola frondosa (Disks.) on mice.【Method】According to the polarity from low to high,petroleum ether,ethyl acetate,acetone and methanol were heated for reflux extraction of Grifola frondosa fruiting body to obtain different extracts.A total of 70 SPF Kunming mice (half male and half female) were divided into 7 groups with 10 in each group,which were marked as normal group,model group,CTX group and Grifola frondosa petroleum ether group,ethyl acetate group,acetone group and methanol extract group,respectively.The mice were given medicine starting on the second day after tumor inoculation.The normal group was given no medicine and saline,the model group was given physiological saline,the CTX group was intraperitoneally injected with CTX 20 mg/kg every other day,while the petroleum ether group,ethyl acetate group,acetone group and methanol extract group were filled with the corresponding extracts of Grifola frondosa every day for 10 days.The tumor inhibition rate,organ index and changes of interferon-γ (IFN-γ),interleukin-2 (IL-2),vascular endothelial growth factor (VEGF),cataalase (CAT),malondialdehyde (MDA) and superoxide dismutase (SOD) in serum were determined and tumor slices were observed to analyze the anti tumor activities,immune regulation effects and anti-oxidation activities of different extracts from Grifola frondosa fruiting body on Hep-A-22 tumor bearing mice.The immune competence of Grifola frondosa extracts was further studied by mice ear edema and mice peritoneal macrophage phagocytosis.【Result】 All groups showed inhibitory effects on tumor growth,and the methanol extract had the best effect (52.31%) with significant difference (P<0.01) with the model experimental group.The contents of IL-2,IFN-γ and SOD in serum of the treated group were very significantly increased (P<0.01) and CAT were significantly increased (P<0.05),showing correlation with the tumor inhibition rate.Increased necrotic area of tumor was observed by HE staining.All groups showed inhibitory effect on the swelling of xylene in mice and methanol group and ethyl acetate group had better effect,which was significantly different from that of normal group (P<0.01).【Conclusion】Methanol and ethyl acetate extracts of Grifola frondosa had good inhibitory effect on tumor growth.Methanol group significantly increased the peritoneal macrophage activity in mice and improved the immune activity of the body,which was related with its anti-tumor effect.
