板栗抗寒性相关指标筛选与评价方法建立

摘要:
【目的】建立板栗抗寒性的鉴定评价方法,筛选可用于板栗抗寒性鉴定的形态和生理指标。【方法】以源自8个省份的16个板栗品种(系)休眠期1年生枝条为试验材料,采用电导法、氯化三苯基四氮唑(TTC)染色法和组织褐变法,配合Logistic 方程确定枝条的低温半致死温度( LT50),分析3种方法确定的LT50之间的相关性,以及板栗枝条各生理指标和组织结构与LT50的相关性。【结果】16个板栗品种(系)枝条相对电导率均呈近似“S”型变化趋势,基于相对电导率拟合的LT50差异较大,其值为-26.64~-17.73 ℃。16个板栗品种(系)枝条TTC 染色的总着色度随处理温度下降呈倒“S”型曲线变化,而枝条冻害指数和芽褐变率均随温度下降呈“S”型曲线变化,基于这3个指标拟合的LT50基本一致,且与基于相对电导率拟合的LT50接近;电导法、组织褐变法及 TTC 染色法确定的16种板栗的LT50之间呈极显著正相关;板栗枝条木栓层厚度与木栓层比率与LT50呈极显著负相关,-20 ℃冷冻处理下的相对电导率和丙二醛含量与LT50呈极显著正相关,可作为抗寒性鉴定的辅助形态和生理指标。【结论】电导法、组织褐变法及 TTC 染色法均可用于休眠期板栗树种的抗寒性鉴定,且以电导法测定的板栗抗寒性结果最为准确。

Abstract:
【Objective】This study established an accurate and reliable method for identifying cold resistance of chestnut to screen the morphological and physiological indexes for identifying cold resistance of chestnut.【Method】The test materials were annual dormant branches of 16 chestnut varieties (strains) from 8 provinces.The semi lethal temperatures (LT50) were assessed by conductometry,triphenyl tetrazolium chloride (TTC) dyeing and tissue browning method with Logistic equation separately.The correlations of LT50 assessed by 3 methods were analyzed,and the correlations between LT50 and physiological index or organizational structure were analyzed.【Result】The relative electrical conductivities of the 16 chestnut varieties (strains) were in “S” trend approximately, and the LT50 based on the relative conductivity were different,ranging from -26.64 to -17.73 ℃.The dyeing degree of the 16 chestnut varieties (strains) were in inverted “S” trend with the decrease of treatment temperature,while the branch freeze injury index and the bud browning rate were in “S” trend.The LT50 based on the three indicators were basically identical,and were close to LT50 based on the relative electrical conductivities.LT50 assessed by conductometry,tissue browning and triphenyl tetrazolium chloride (TTC) dyeing method had significant positive correlations.The significant negative correlation was obtained between LT50 and phellem layer thickness or phellem layer ratio in the branch,and there was significant positive correlation between LT50 and relative electrical conductivities (REC) or malonaldehyde content at -20 ℃.The phellem layer thickness or phellem layer ratio in the branch could be used as morphological indexes,and REC and malonaldehyde content of branch at -20 ℃ could be used as physicochemical indexes.【Conclusion】Conductometry,tissue browning and TTC dyeing method could be used to identify the cold resistance of chestnut in dormant period.The conductometry method was the best.
