神经激肽B在犬生殖轴上的表达

摘要:
【目的】研究神经激肽B(NKB)在犬生殖轴上的表达规律。【方法】以仔犬期、幼犬期、初情期母犬及成年公犬、母犬为研究对象,颈动脉放血处死后取其下丘脑、垂体、睾丸和卵巢,其中成年公、母犬的样本进行NKB免疫组化试验,观察NKB分布情况并拍照;仔犬期、幼犬期、初情期和成年期母犬样本进行RT-qPCR试验,以GAPDH为内参,测定样本中目的基因Tac3 mRNA的表达量。【结果】NKB主要分布于下丘脑外侧区和视上核、腺垂体、卵巢卵泡颗粒层细胞和卵母细胞及睾丸的间质组织和生精细胞,而神经垂体中未见分布。在母犬下丘脑、垂体和卵巢中,Tac3 mRNA表达量均表现为从仔犬期至初情期逐渐上升,于初情期达最高水平,至成年期又有所下降,且与其他各发育阶段差异显著(P<0.05)。【结论】NKB可能参与母犬初情期的启动。

Abstract:
【Objective】This study analyzed the expression pattern of neurokinin B (NKB) in the reproductive axis of dogs.【Method】Infancy,puppy and puberty female dogs as well as adult male and female dogs were anesthetized and bled to death via cutting the carotid artery.The hypothalamus,pituitary,testis and ovaries were collected.The samples from adult male and female dogs were used for immunohistochemistry experiment and distribution of NKB was observed and captured.The samples from infancy,puppy,puberty and adulthood female dogs were used for RT-qPCR to detect the expression levels of Tac3 mRNA using GAPDH as the reference gene.【Result】NKB immune positive cells were mainly distributed in the lateral hypothalamic area (LHA) and supraoptic nucleus (SON) of hypothalamus,adenohypophysis, granular layer and oocytes of ovary,interstitial tissue and spermatogenic cells of testis.There was no positive cells in neurohypophysis.The levels of Tac3 mRNA in hypothalamus, pituitary and ovary were increased from infancy to puberty stage,then decreased at adulthood stage with significant difference (P<0.05).【Conclusion】NKB may participate in the puberty launching of female dogs.
